瓜蒌薤白汤含药血清对博来霉素诱导的大鼠肺泡巨噬细胞纤维化的影响

摘　　要：目的:观察瓜蒌薤白汤含药血清作用于博来霉素诱导的NR8383细胞功能变化,探讨瓜蒌薤白汤抗纤维化作用与细胞外信号调节激酶1/2(extracellular signal-regulated kinase 1/2,ERK1/2)通路的关系。方法:实验分为正常对照组、博来霉素组、地塞米松组、空白对照血清低体积分数组、瓜蒌薤白汤含药血清低体积分数组、空白对照血清中体积分数组、瓜蒌薤白汤含药血清中体积分数组、空白对照血清高体积分数组、瓜蒌薤白汤含药血清高体积分数组。酶联免疫吸附法检测细胞上清液中转化生长因子β1(transforming growth factor-β1,TGF-β1)、Ⅰ型胶原(collagen type-Ⅰ,COL-Ⅰ)、Ⅲ型胶原(collagen type-Ⅲ,COL-Ⅲ)、蛋白质免疫印迹法检测ERK与p-ERK蛋白表达情况。结果:博来霉素组TGF-β1、COL-Ⅰ、COL-Ⅲ表达均增加,不同体积分数瓜蒌薤白汤含药血清干预后,以上指标表达量均下降,中体积分数效果最好(P<0.05);蛋白质免疫印迹法结果显示,博来霉素组p-ERK1/2的表达水平均升高,不同体积分数瓜蒌薤白汤含药血清干预后,p-ERK1/2的表达水平均降低,中体积分数效果最好(P<0.05)。结论:瓜蒌薤白汤含药血清可通过ERK1/2信号通路降低TGF-β1和COL-Ⅰ、COL-Ⅲ蛋白表达,这可能是瓜蒌薤白汤治疗肺纤维化的作用机制之一。Objective: To observe the changes in the function of bleomycin-induced NR8383 cells in Gualou Xiebai Decoction serum,and to explore the anti-fibrotic effect and extracellular signal-regulated kinase 1/2( ERK1/2) pathway correlation. Methods:The experiment was divided into normal control group,bleomycin group,dexamethasone group,blank control serum low volume fraction array,Gualou Xiebai Decoction serum low volume fraction array,blank control serum volume fraction array,and Diaolou The volume fraction array of Zhibai Decoction-containing serum,the high-volume fraction array of blank control serum,and the high-volume array of Gualou Xiebai Decoction-containing serum. Enzyme-linked immunosorbent assay for the detection of transforming growth factor-β1( TGF-β1),collagen type-Ⅰ( COL-Ⅰ),and collagen type-Ⅲ in cell supernatants. (COL-Ⅲ) and Western blotting were used to detect ERK and p-ERK protein expression. Results: The expressions of TGF-β1,COL-Ⅰ,and COL-Ⅲ in the bleomycin group were increased. After intervention with different volume fractions of the Gualou Xiebai Decoction-containing serum,the expressions of the above indicators all decreased,and the medium volume fraction had the best effect( P < 0. 05);Western blot analysis showed that the expression level of p-ERK1/2 in the bleomycin group was increased,and the expression level of pERK1/2 was increased after intervention with different volume fractions of Gualou Xiebai Decoction-containing serum There was a decrease,and the effect of medium volume fraction was the best( P < 0. 05). Conclusion: The drug-containing serum of Gualou Xiebai Decoction can reduce the expression of TGF-β1,COL-Ⅰ and COL-Ⅲ through the ERK1/2 signaling pathway,which may be one of the mechanisms of the action of Gualou Xiebai Decoction in treatment of pulmonary fibrosis.
