真武汤和越婢汤对阿霉素肾病大鼠AQP1/AQP2的影响

摘　　要：目的:分别观察真武汤、越婢汤对阿霉素肾病大鼠肾组织AQP1、AQP2表达的影响,以探讨中医不同利水法的作用机制及差异。方法:选用雄性SD大鼠48只按随机数字表法分为空白组、模型组、真武组及越婢组,采用尾静脉注射阿霉素建立肾病模型,药物灌胃6周后生化检测各组大鼠24 h尿蛋白定量、肝功(ALB,ALT)、肾功(Scr,BUN)及血脂(CHOL,TG)等,HE染色观察大鼠肾脏病理,免疫组化检测肾组织AQP1、AQP2表达。结果:与模型组比较中药2组24 h尿蛋白定量、血脂均明显下降,白蛋白升高,肾脏病理均明显改善,肾组织AQP1、AQP2表达均明显升高,但中药2组比较差异无统计学意义。结论:真武汤和越婢汤均可上调阿霉素肾病大鼠肾组织AQP1、AQP2的表达,改善上述生化指标,减轻肾脏病理损害,但中药2组之间比较差异无统计学意义。Objective:To investigate the effects of Zhenwu Decoction and Yuebi Decoction on the expression of AQP1/AQP2 in the kidney of adriamycin-induced nephropathy rats,and to explore the mechanism and difference of different diuretic methods of traditional Chinese medicine(TCM).Methods:48 male SD rats randomly divided into four groups:blank group,model group,Zhenwu and Yuebi Decoction.The model of adriamycin-induced nephropathy was established by tail vein injection.After six weeks,24h urinary protein,ALB,ALT,Scr,BUN,CHOL and TG were measured in rats of each group.HE staining was used to observe the pathological changes of rat kidney.Immunohistochemistry was used to detect the expression of AQP1,AQP2.Results:Compared with the model group,the 24h urinary protein and blood lipids in the two groups of Chinese medicine decreased significantly,and albumin increased;renal pathology was obviously improved;the expression of AQP1,AQP2 in renal tissue was significantly increased.But there was no difference in the two Chinese medicine group.Conclusion:Zhenwu and Yuebi Decoction can both up-regulate the expression of AQP1,AQP2 in renal tissue of adriamycin nephropathy rats,improve the above biochemical indexes and alleviate the pathological damage of kidney,but there was no difference between the two groups.
