摘 要:目的优化四妙凉血颗粒的提取工艺参数。方法以得膏率和盐酸小檗碱转移率为评价指标,采用单因素法筛选溶剂倍数、提取时间对评价指标的影响。以总评归一OD值为指标,采用星点设计法设计实验,通过效应面法优化提取工艺。结果预测得到最佳的二次多项回归方程为:Y=-0.456+0.0688A+8.64×10-3B-6×10-5AB-1.57×10-3A2-2.5×10-5B2,拟合度为0.9547。最佳提取参数为:溶剂倍数19倍,提取时间150 min。经3批验证实验:盐酸小檗碱的转移率、得膏率分别为为20.95%±1.7%、20.54%±0.47%,实际OD值与预测的OD值偏差为-5.94%。结论星点设计效应面法可用于四妙凉血颗粒提取工艺的优化,所建模型预测准确,优化后的提取工艺稳定、可靠。OBJECTIVE To optimize the extraction process parameters of Simiao Liangxue granules.METHODS The dry extact yield rate and the berberine hydrochloride transfer rate were evaluated.The single factor method was used to screen the solvent fold and the extraction time on the index.Based on the OD value,the design method was designed by central composite design,and the extraction process was optimized by the response surface methodology.RESULTS The best quadratic multiple regression equation predicted was Y=-0.456+0.0688A+8.64×10-3B-6×10-5AB-1.57×10-3A2-2.5×10-5B2,and the fitness was 0.9547.The optimal extraction parameters after optimization were the follow:solvent fold was 19 fold,and extraction time was 150 min.After three batches of verification experiments,the berberine hydrochloride transfer rate and the obtained paste rate were 20.95%±1.7% and 20.54%±0.47%,respectively,and the deviation between the actual OD value and the predicted OD value was-5.94%.CONCLUSION The central composite design/response surface methodology can be used to optimize the extraction process of Simiao Liangxue granules.The established model is accurate and the optimized extraction process is stable and reliable.
