摘  要:目的研究芫根总多糖的抗氧化作用以及对模拟高原缺氧小鼠脑、肺的保护作用。方法采用低压低氧舱模拟海拔4000 m的高原环境,评估芫根总多糖的抗高原缺氧活性。结果连续7 d低氧暴露后,与常压常氧组比较,低压低氧模型组小鼠脑组织海马区、肺组织出现明显的病理损伤,脑、肺组织中的丙二醛(MDA)、乳酸(LD)的浓度和乳酸脱氢酶(LDH)的活性显著升高,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)、ATP酶(ATPase)的活性显著降低。经过芫根总多糖给药处理后,高剂量组小鼠脑组织海马区、肺组织的病理损伤得到缓解,脑、肺组织中MDA、LDH和LD的活性显著降低,而SOD、GSH-Px、T-Aoc、ATPase的活性显著升高。结论芫根总多糖具有抗高原缺氧的作用,能保护模拟高原环境中缺氧小鼠的脑组织和肺组织,可能主要是通过升高机体自身抗氧化能力、降低氧自由基损伤、调节自身能量代谢来发挥抗高原缺氧的作用。OBJECTIVE To study the antioxidant effect of total polysaccharides of Brassica rapa L.and the protective effect on brain and lung of mice with simulated altitude hypoxia.METHODS The anti-hypoxia activity of total polysaccharides of B. rapa was evaluated by simulating the altitude environment of 4000 meters with hypobaric hypoxic experimental chamber.RESULTS The results of experiment showed that during the 7 days of continuous hypoxia by simulating an altitude of 4000 m,compared with the normal group,model mice in the hypobaric oxygen chamber has a significant pathological tissue damage in lung and hippocampus of brain.The concentration of malondialdehyde(MDA) and lactic acid(LD),and the activity of lactate dehydrogenase(LDH) were increased significantly.The activities of superoxide dismutase(SOD),glutathion peroxidase(GSH-Px),total antioxidant capacity(T-Aoc) and ATP synthase(ATPase) were decreased dramatically.However,after the treatment with total polysaccharides of B. rapa,the pathological damage degree of hippocampal area and lung of the mice in the high dose group was reduced,the concentration of MDA and LD as well as the activity of LDH in brain and lung tissue were significantly decreased,and the activity of SOD,GSH-Px,T-Aoc and ATPase were significantly increased.CONCLUSION The total polysaccharides of B. rapa have the function of antioxidant activity,which can protect the brain and lung tissues of hypoxic mice in a simulated high altitude environment.
