摘  要:目的探讨氢分子对高脂饮食C57BL/6小鼠肝脏的保护作用及其分子机制。方法给予50只4周龄C57BL/6小鼠高脂饮食,适应喂养1周后剔除体重增长不明显的小鼠,选取平均体重增长率一致的20只小鼠依据随机数字法分为富氢水组和对照组,每组10只。两组小鼠继续给予高脂饮食并分别腹腔注射富氢水[10 mL/(kg·d)]及0.9%氯化钠溶液[10 mL/(kg·d)],共计8周。检测两组小鼠血浆中血脂[总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)]、氧化指标[超氧化物歧化酶(SOD)、丙二醛(MDA)]的水平;应用苏木精-伊红染色法(HE)观察肝细胞形态学变化;采用酶联免疫吸附试验法检测小鼠肝脏组织炎症因子[白细胞介素(IL)-6、IL-8、IL-10、肿瘤坏死因子-α(TNF-α)]蛋白的表达。结果富氢水组小鼠体重增长率低于对照组[(0.73±0.06)%比(1.06±0.09)%](P<0.001)。富氢水组TC、TG、LDL-C水平低于对照组[(3.11±0.34)mmol/L比(4.61±0.79)mmol/L,(0.92±0.23)mmol/L比(1.13±0.20)mmol/L,(3.07±0.46)mmol/L比(4.45±0.86)mmol/L](P<0.05),HDL-C水平高于对照组[(6.74±0.96)mmol/L比(3.71±0.30)mmol/L](P<0.001)。富氢水组MDA低于对照组[(0.71±0.05)nmol/mgprot比(1.02±0.10)nmol/mgprot],SOD高于对照组[(347.73±12.02)U/mgprot比(268.78±12.90)U/mgprot](P<0.001)。HE染色结果显示,富氢水组小鼠肝小叶结构完整,肝索排列整齐,肝细胞未有明显损伤,核结构非常清晰,肝脏组织中有少许小脂滴;对照组小鼠肝小叶结构被破坏,肝索排列紊乱,肝细胞体积明显增大,细胞中有大脂滴出现,呈空泡状,占据整个细胞,并将细胞核挤向一侧。富氢水组IL-6、IL-8、TNF-α低于对照组,IL-10高于对照组(P<0.05)。结论氢分子能够调节高脂饮食小鼠血脂水平,抑制肝脏氧化应激及炎症反应,减少肝脏脂肪沉积。Objective To investigate the protective effects of hydrogen molecules on the liver of C57BL/6 mice fed with a high fat diet and its molecules mechanisms.Methods Fifty 4-week-old C57BL/6 mice were given a high-fat diet,and mice with insignificant weight gain were eliminated after one week of adaptive feeding.20 mice with the same average weight growth rate were selected and divided into a hydrogen-rich water group and a control group according to the random number method,10 in each group.Both groups were given high fat diet,and intraperitoneal injection of hydrogen rich water[10 mL/(kg·d)]and normal saline[10 mL/(kg·d)]respectively,for a total of 8 weeks.The plasma levels of total cholesterol(TC),triglycerides(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),oxidation index malondialdehyde(MDA)and superoxide dismutase(SOD)of the two groups were tested.The morphological changes of liver cells were observed via hematoxylin and eosin staining(HE)staining;enzyme-linked immunosorbent assay method was used to detect the protein expression of inflammation factors,such as tumor necrosis factor-α(TNF-α),interleukin(IL)-6,IL-8 and IL-10,in mice liver tissue.Results The weight growth rate of the hydrogen-rich water group was lower than that of the control group[(0.73±0.06)%vs(1.06±0.09)%](P<0.001).The levels of TC,TG,LDL-C of the hydrogen-rich water group were lower than those of the control group[(3.11±0.34)mmol/L vs(4.61±0.79)mmol/L,(0.92±0.23)mmol/L vs(1.13±0.20)mmol/L,(3.07±0.46)mmol/L vs(4.45±0.86)mmol/L](P<0.05),the HDL-C was higher than that of the control group[(6.74±0.96)mmol/L vs(3.71±0.30)mmol/L](P<0.001).The MDA of the hydrogen-rich water group was lower than that of the control group[(0.71±0.05)nmol/mgprot vs(1.02±0.10)nmol/mgprot],SOD higher than that of the control group[(347.73±12.02)U/mgprot vs(268.78±12.90)U/mgprot](P<0.001).The results of HE staining showed that in the hydrogen-rich water group the structure of hepatic lobule was complete,the hepa
