NLRP3在运动大鼠骨骼肌中的表达及干预研究

摘  要:随机将24只雄性SD大鼠分为:安静对照组(C)、运动损伤组(M)、自然恢复组(N)、草珊瑚干预组(S)四组。采用连续下坡跑(四周),跑台坡度-16°,5 d/w。第1周,速度16 m/min,每日60 min;第2-3周,采用速度20 m/min,60 min/d;第4周,周一至周四,采用速度20 m/min,90 min/d,周五速度25 m/min,时间100 min。安静对照组(C)与运动损伤组(M)大鼠在最后一次离心运动结束24 h后,腹主动脉采血和取材;自然恢复组(N)采用自然恢复7 d,草珊瑚干预组(S)灌胃给药草珊瑚粗多糖7 d,之后过夜禁食12 h后腹主动脉采血和取材。结果表明:离心运动24 h后(M组)骨骼肌细胞结构发生损伤性变化,可见肌节排列紊乱,出现Z线流、Z线波形变化等,N组可见有一定修复与改善,S组则基本修复,肌节排列、Z线结构基本正常;NLRP3、IL-1β、IL-18蛋白表达在大鼠大强度离心运动运动24 h后显著性升高,自然修复7 d后仍然保持较高水平。草珊瑚粗多糖能够有效抑制大强度离心运动后大鼠骨骼肌相关炎性因子的表达水平,显现出草珊瑚粗多糖能够有效抑制炎性因子,改善炎症反应过程。Twenty-four male Wistar rats were randomly divided into four groups,which were Control group(C),Muscle injury group(M),Natural recovery group(N,)and Sarcandra glabra intervention group(S).The motion model adopted the continuous downhill running centrifugal movement on all sides with the treadmill slope as-16 degrees,five days a week.The first week was at the speed of 16 m/min for 60 minutes;the second and the third week kept the speed of 20 m/min for 60 minutes;in the fourth week,the first four days were at the speed of 20 m/min for 90 minutes,and the fifth day changed the speed to 25 m/min and lasted for 100 minutes.The experiment was based on the following samples:the blood samples of C group and M group rats were collected from abdominal aorta after twenty four hours of rats’last heavy load eccentric exercise;the samples of N group took nature repair 7 d;the blood samples of S group rats were collected from abdominal aorta after overnight fasting for twelve hours by intragastric administration of Sarcandra glabra 7 d.The reesults are as follows:After twenty four hours of eccentric exercise,the structure of skeletal muscle cells in M group underwent injurious changes;the sarcomas were arranged in disorder;there were Z-line flow and Z-line waveform changes,and so on.N group could see some repair and improvement and S group was basically repaired and the sarcomas arrangement and Z-line structure were basically normal.NLRP3,IL-1β,IL-18 protein expression was significantly increased and nature repair 7 d remained at a high level after twenty four hours of rats’heavy load eccentric exercise.The conclusion showed that polysaccharides of Sarcandra glabra could effectively inhibit the expression level of related inflammatory factors,which showed that Sarcandra glabra was effective in inhibiting inflammatory factors and improving the inflammatory processes.
