摘  要:目的研究谷氨酸AMPA/Kainate受体拮抗剂CNQX对断尾诱发小鼠前扣带回皮质神经元c-fos基因表达的影响。方法免疫组化方法分别研究尾静脉注射和蛛网膜下腔注射CNQX对尾末端2.5 cm剪断后0.5 h小鼠前扣带回皮质神经元c-fos基因表达的影响。结果正常小鼠前扣带回皮质神经元有少量c-fos基因表达,而断尾后0.5 h小鼠前扣带回皮质神经元c-fos基因表达显著增强。同时,尾静脉注射和蛛网膜下腔注射CNQX均能拮抗该c-fos基因表达的显著增强。结论断尾后小鼠前扣带回皮质神经元c-fos基因表达显著增强的过程有外周与中枢谷氨酸AMPA/Kainate受体参与。Objective We aimed to study the effect of glutamate AMPA/kainate receptor antagonist CNQX on tail amputation induced c-fos gene expression in the anterior cingulate cortex neurons of mice.Methods CNQX was injected into the caudal vein or subarachnoid space.The effect of CNQX on gene expression at 0.5 h after cutting 2.5 cm of the tail extremity was studied using immunohistochemistry.Results In the control group,a small amount of c-fos gene was expressed in the anterior cingulate cortex neurons.However,the expression of c-fos in the anterior cingulate cortex neurons of mice increased significantly 0.5 h after tail amputation;this increase was antagonized by CNQX injection into the caudal vein or subarachnoid space.Conclusion Glutamate peripheral and central AMPA/kainate receptors are involved in tail amputation induced increase in c-fos gene expression in the anterior cingulate cortex neurons of mice.
