摘 要:目的:研究非同源末端连接修复基因Ligase4缺失对芹菜素(APG)抑制TK6细胞增殖作用的影响。方法:采用CCK8方法检测APG和阳性药依托泊苷(ETP)对TK6细胞增殖的影响;采用流式细胞术检测APG对TK6细胞周期和凋亡的影响;利用免疫荧光实验分析APG对野生型(WT)和DNA损伤修复缺陷的Ligase4-/-细胞的DNA损伤作用。结果:APG对TK6细胞的增殖抑制作用呈现剂量依赖性;APG能将TK6细胞阻滞在G2/M期并诱导TK6细胞凋亡;经80μmol·L^-1APG处理细胞后,Ligase4-/-型细胞的γ-H2AX foci相对于WT细胞明显增加,在6 h达到峰值(P<0. 05)。结论:APG能抑制TK6细胞的增殖,并且对非同源末端连接修复基因Ligase4缺失的细胞造成的DNA损伤更严重且抑制细胞增殖作用更强。Objective: To study the effect of deficiency of non-homologous terminal junction repair gene Ligase4 on the inhibition of apigenin(APG)on TK6 cell proliferation. Methods: CCK8 method was used to detect the effects of APG and positive drug etoposide(ETP)on the TK6 cell proliferation. Flow cytometry was used to detect the effects of APG on TK6 cell cycle and apoptosis. Immunofluorescence assay was used to analyze the DNA damage effect of APG on wild-type(WT)and Ligase4-/-cells with DNA damage repair defects.Results: The inhibitory effect of APG on TK6 cell proliferation was dose-dependent. APG could block TK6 cells in G2/M phase and induce TK6 cells apoptosis. After treatment with 80 μmol·L^-1 APG,the γ-H2 AX foci of Ligase4-/-cells was significantly increased compared with that of WT cells with a peaked at 6 h(P<0. 05).Conclusion: APG can inhibit the TK6 cell proliferation,and can cause more serious DNA damage and inhibit cell proliferation in the cells with Ligase4 deficiency of non-homologous terminal ligation junction gene.
