摘  要:目的:观察巨噬细胞极化在七氟烷致大鼠认知功能障碍中的作用。方法:选取健康SPF级5个月龄Wistar大鼠60只,按随机数字表法将实验大鼠分为对照组(n=30)和观察组(七氟烷麻醉处理,n=30)。对照组大鼠不开启七氟烷挥发罐,吸入纯氧,氧气流量为2 L/min,时间2 h;观察组大鼠氧气流量为2 L/min,同时调节七氟烷挥发罐维持麻醉箱气体出口七氟烷浓度为2%,时间2 h。分别在麻醉前一天(T0)、麻醉后第5天(T1)、第10天(T2)、第20天(T3)和第30天(T4)进行水迷宫测试,观察并记录逃避潜伏期(ELP);每个时间点(T0~T4)水迷宫测试结束后,两组各取大鼠5只,采用流式细胞仪检测M1型和M2型巨噬细胞占比和用酶联免疫吸附测试(ELISA)法测定大鼠海马白介素(IL)-8和IL-10表达情况。结果:观察组大鼠T1和T2时刻ELP较T0时刻显著延长(P<0.05),T3和T4时刻均无明显变化(P>0.05);与对照组比较,观察组大鼠在T1和T2时刻ELP显著延长(P<0.05),T3和T4时刻均无明显变化(P>0.05)。与T0时刻比较,观察组海马T1和T2时刻IL-8表达、M1巨噬细胞占比和M1/M2显著升高(P<0.05),IL-10表达和M2巨噬细胞占比显著降低(P<0.05);T3和T4时刻上述指标均无明显变化(P>0.05)。与对照组比较,观察组大鼠T1和T2时刻IL-8表达、M1巨噬细胞占比和M1/M2显著升高(P<0.05),IL-10表达和M2巨噬细胞占比显著降低(P<0.05);T3和T4时刻上述指标均无明显变化(P>0.05)。结论:七氟烷可能通过影响海马巨噬细胞极化引起海马神经阻滞促炎因子水平升高,从而导致大鼠术后短期认知功能障碍。Objective:To observe the role of macrophage polarization in sevoflurane-induced cognitive dysfunction in rats.Methods:Sixty healthy 5-month-old SPF Wistar rats were selected and divided into control group(n=15)and observation group(sevoflurane anesthesia treatment,n=15)using a random number table.Both groups were placed in the anesthesia box for 2 h.The rats in the control group inhaled pure oxygen at a flow rate of 2 L/min;in the observation group,the oxygen flow rate was 2 L/min,and the sevoflurane vaporizer was adjusted to maintain the concentration of sevoflurane at the gas outlet of the anesthesia box at 2%.Water maze tests were performed on the day before anesthesia(T0)and the 5 th day(T1),10 th day(T2),20 th day(T3),and 30 th day(T4)after anesthesia,and the escape latency period(ELP)was recorded.After each water maze test(T0-T4),five rats were taken from each group,and the proportion of M1 and M2 macrophages was detected by flow cytometry,while the expression level of hippocampus interleukin(IL)-8 and IL-10 was detected using enzyme-linked immunosorbent assay(ELISA).Results:In observation group,the ELP at T1 and T2 were significantly longer than that at T0(P<0.05),and there was no significant change at T3 and T4(P>0.05).Compared with the control group,the ELP of observation group at T1 and T2 were significantly longer(P<0.05),and there was no significant change at T3 and T4(P>0.05).In the observation group,the expression of IL-8,the proportion of M1 macrophages,and M1/M2 in the hippocampus were significantly increased(P<0.05),while the expression of IL-10 and the proportion of M2 macrophages significantly decreased at the time of T1 and T2(P<0.05)when compared to those at T0;there was no significant change in the above indexes at T3 and T4(P>0.05).Compared with the control group,the expression of IL-8,the proportion of M1 macrophages and M1/M2 were significantly increased(P<0.05),and the expression of IL-10 and the proportion of M2 macrophages were significantly reduced(P<0.05)at T1 and T2 in the observati
