多浪羊NCOA1基因多态性与产羔数的相关分析

摘　　要：分析研究了新疆多浪羊核受体辅激活蛋白基因(Nuclear receptor coactivator1 gene,NCOA1)的多态性及其与产羔数的相关性。结果表明:NCOA1基因存在两个多态位点,分别为内含子12的g.166397 A>C突变位点和外显子17的g.187501 G>A突变位点;g.166397 A>C突变位点的优势基因为A等位基因,基因频率为0.968;g.187501 G>A突变位点的优势基因为B等位基因,基因频率为0.643,且在该突变位点发生了错义突变,由原编码的丝氨酸(Ser)改变为谷酰胺酸(Gln);在多浪羊NCOA1基因的g.166397 A>C突变位点,AA基因型个体的产羔数显著高于BB基因型个体的(P<0.05);在多浪羊NCOA1基因的g.187501 G>A突变位点,BB基因型个体的产羔数显著高于AA和AB基因型个体的(P<0.05)。由此推断NCOA1基因可以作为多浪羊多羔性状的候选基因。The correlation between NCOA1(Nuclear receptor coactivator 1)gene polymorphism and fertility trait of Duolang sheep was analyzed.The results showed that there were two polymorphic loci in NCOA1 gene,g.166397 A>C mutation in intron 12 and 17 and g.187501 G>A mutation in exon 12.The dominant gene of g.166397 A>C mutation loci was A allele with a gene frequency of 0.968.The g.187501 G>A mutation loci was B allele with a gene frequency of 0.643,and the mutation site was missense mutation,which was changed from the original serine(Ser)to glutamic acid(Gln).The correlation analysis between two SNP loci of NCOA1 gene and litter size showed that the litter size of AA genotype of NCOA1 gene was significantly higher than that of BB genotype(P<0.05).The litter size of BB genotype of 187501 G>A genotype of NCOA1 gene was significantly high?er than that of AA and AB genotype(P<0.05).It could be concluded that NCOA1 gene could be used as a candidate gene for the multi-lamb trait of Duolang sheep.
