摘 要:[目的]研究深海网箱养殖系统许氏平鲉(Sebastes schlegeli)肠道与环境中菌群结构的周年变化规律及其与疾病发生的相关性。[方法]采用高通量测序技术在2016—2017年一个年度内对同一网箱中不同月份许氏平鲉的肠道(Int)、海水(W)、冰鲜饵料(F)及网衣附着物(N)中的细菌多样性及群落结构进行比较分析。[结果]不同月份深海网箱养殖系统中菌群多样性为W>N>F>Int。温度和季节变化可影响养殖系统中前20种优势菌种丰度的排名次序,其中肠道与冰鲜饵料样品中发光杆菌属(Photobacterium)在2016年7、11月出现较大比重,对应的OTU数量占该样品全部OTU数量的28.07%、19.25%和8.99%、38.75%。同时,肠道与冰鲜饵料样品中弧菌属(Vibrio)在2016年8、9月,2017年6、8月样品中相对丰度显著上升,分别为5.72%、5.94%、8.40%和8.34%、6.08%、30.73%。PCoA图表明,加权与未加权条件下水样和网衣附着物样品菌群相关性较低,而肠道样品与冰鲜饵料菌样品群相关性较高。[结论]许氏平鲉深海网箱养殖系统周年内在温度较高的月份致病菌比例明显增加,冰鲜饵料作为病原菌主要载体,是影响肠道菌群结构的主要因素,成为网箱养殖系统鱼类发病的潜在隐患。[Objective]To study the annual change of intestinal and environmental flora structure of Sebastes schlegeli in the deep-sea cage culture system and its correlation with disease occurrence.[Method]The high-throughput sequencing technology was used to compare and analyze the bacterial diversity and community structure in the intestine(Int),culture water(W),chilled bait(F)and net attachment(N)of S.schlegeli in the same cage in different months from 2016 to 2017.[Result]The flora diversity in deep-sea cage culture system in different months was W>N>F>Int.As the temperature and season changes,the order of the top 20 dominant species abundance in the survival system can be affected.Among the Int and F samples,the Photobacterium in the July and November 2016 samples had a large proportion.The corresponding number of OTUs accounted for 28.07%,19.25%and 8.99%,38.75%of the total OTU of the sample.At the same time,the relative abundance ratio of Vibrio in the Int and F samples in August,September,2016 and June,August 2017 were significantly increased,which was 5.72%,5.94%,8.40%and 8.34%,6.08%,30.73%,respectively.PCoA showed that under the weighted and unweighted conditions,the correlation between W and N was relatively small,while the correlation between Int and F was relatively large.[Conclusion]The percentage of pathogenic bacteria increased significantly in the month of higher temperature during the anniversary of S.schlegeli deep-sea cage culture system.As a main carrier of pathogenic bacteria,chilled bait was the main factor affecting the structure of intestinal flora and became potential risk of fish disease in cage culture system.
