摘 要:目的探索聚葵二酸甘油酯(PGS)材料用于构建特定形状组织工程脂肪的可行性。方法将PGS通过盐析法制备成三维多孔支架。电镜观察PGS支架的表征;体外细胞黏附实验观察PGS支架的细胞相容性;将处理后的脂肪组织与支架复合,植入裸鼠皮下,4周后取材行大体观察和组织学检测。结果SEM观察显示,ADSCs接种PGS支架后7 d,支架材料上可见细胞黏附并大量增殖,显示出良好的细胞相容性;体内培养4周后取材,大体观察显示支架高度及直径维持良好,HE染色显示支架周围有脂肪细胞浸润生长,植入物内有炎性细胞浸润,Masson染色显示植入物周围包裹了一层纤维包膜,支架侧面脂肪细胞浸润面积明显多于底部。结论PGS支架对于构建特定外形脂肪组织具有潜在应用价值。Objective To explore the feasibility of poly(glycerol sebacate)(PGS)material for constructing tissue-engineered adipose with specific shape.Methods 3D porous PGS scaffold was prepared by salting out method.The characterization of PGS scaffold was observed by scanning electron microscopy(SEM).The cytocompatibility of PGS was observed by cell adhesion test.The fat treatment was compounded with scaffolds and implanted subcutaneously in nude mice.Gross observation and histological examination were performed 4 weeks later.Results SEM observation showed that ADSCs were adhered to PGS scaffolds and proliferated within scaffolds after seeding for 7 days,indicating good cytocompatibility of PGS scaffold.After 4 weeks of in vivo culture,general observation showed that the height and diameter of scaffolds were well maintained.HE staining revealed the infiltration and growth of adipocytes around the scaffolds as well as the recruitment of inflammatory cells inside the implants.Masson staining showed that the implant was encapsulated by a fibrous capsule,and the infiltration area of adipocytes on the side of the scaffold was significantly larger than that at the bottom.Conclusion PGS scaffold has potential application value for constructing specific-shape adipose tissue.
