摘  要:目的比较分析样本放置时间对流式细胞术检测单核细胞亚群的检测效果。方法随机法选择20例本院健康体检者,分别采用FFS-SSC、CD45和CD14的设门方法对单核细胞亚群进行检测,25℃分析样本放置时间对检测结果的影响。结果(1)样本放置8小时与1小时内相比较,FSC-SSC的设门方法、CD14设门方法检测的Mon2群细胞显著上升(P<0.05),Mon3群细胞显著下降(P<0.05);而CD45设门方法,随着时间的延长,Mon1群细胞显著上升(P<0.05),Mon3细胞显著下降(P<0.05)。8小时与4小时内比较,FSC-SSC设门时Mon2显著升高(P<0.05),而CD45设门、CD14设门检测Mon3细胞显著下降(P<0.05)。(2)标本室温条件下,随着放置时间的延长,单核细胞SSC方向检测信号下降。单核细胞出现分群,SSC检测信号较高的群(SSChi)和SSC检测信号较低的群(SSClow)。与样本放置4小时相比,样本放置8小时,SSClow细胞群中Mon1、Mon2、Mon3的比例有显著性差异(P<0.05),而SSChi细胞群中Mon1、Mon2、Mon3的比例无显著性差异(P>0.05)。(3)标本室温条件下放置4小时,SSClow与SSChi相比Mon1、Mon3有显著性差异(t=17.946,P<0.001;t=12.177,P<0.001);室温放置8小时,SSClow与SSChi相比Mon1、Mon2、Mon3均有显著性差异(t=8.278,P<0.001;t=2.864,P=0.019;t=7.323,P<0.001)。结论单核细胞亚群检测分析时易受标本放置时间的影响,标本采集后应尽快检测,采集后4小时内对检测结果影响不大。Objective To comparatively analyze the detection effect of monocyte subgroups by sample placement time in flow cytometry,and to explore the reasons which can provide basis for clinical or scientific research.Meth?ods The level of Monocyte subsets in 20 serum samples from subjects of healthy physical examination was deter?mined by FFS-SSC,CD45 and CD14 gatingmethods respectively.Moreover,the influence of sample placement time was analyzed.Results①Comparing 8 hours with 1 hour,the serum level of Mon2 group cells detected by FSC-SSC and CD14 gatingmethods was increased significantly(P<0.05),and the serum level of Mon3 group cells was decreased significantly(P<0.05).However,with the extension of time,the serum level of Mon1 group cells detected by CD45 gatingmethods was increased significantly(P<0.05),and the serum level of Mon1 group cells was decreased significantly(P<0.05).Comparing 8 hours with 4 hours,the serum level of Mon2 group cells detect?ed by FSC-SSC gatingmethods was increased significantly(P<0.05),and the serum level of Mon3 group cells de?tected by CD45 and CD14 gatingmethods was decreased significantly(P<0.05).②At room temperature,the signal of SSC in mononuclear cells was decreased with the extension of placement time.The mononuclear cells appeared in groups,such as SSC detection signal higher group(SSChi)and SSC detection signal low group(SSClow).Com?paring 8 hours with 4 hours,the ratio of Mon1,Mon2 and Mon3 in SSClow cell group was significantly different(P<0.05),while the ratio of Mon1,Mon2 and Mon3 in SSChi cell group showed no significant difference(P>0.05).③For 4 hours at room temperature,the serum level of Mon1 and Mon3 in SSClow was significantly differ?ent from that in SSChi(t=17.946,P<0.001;t=12.177,P<0.001);For 8 hours at room temperature,the serum lev?el of Mon1、Mon2 and Mon3 in SSClow is significantly different from that in SSChi(t=8.278,P<0.001;t=2.864,P=0.019;t=7.323,P<0.001)Conclusions The detection effect of monocyte subgroups is susceptible to be influenced by sample placem
