摘 要:采用HPLC法测定木鳖子中对羟基苯甲酸含量。色谱条件为:Agela Venusil XBP C18色谱柱(4.6 mm×250 mm,5μm),甲醇-0.1%乙酸为流动相进行梯度洗脱,检测波长254 nm,流速1 mL·min^-1。结果表明,木鳖子中对羟基苯甲酸分离良好,分离度为1.75,理论塔板数为126964;对羟基苯甲酸质量(x)在0.100~0.485μg之间与峰面积(y)线性关系良好,线性方程为y=1000000x-81274(R^2=0.9997);平均加标回收率为107.5%;木鳖子药材中对羟基苯甲酸含量为4.942μg·g^-1。该方法简单、高效,可用于木鳖子中对羟基苯甲酸含量测定。We determined the content of p-hydroxybenzoic acid in Momordica cochinchinensis by HPLC.The chromatographic conditions are determined as follows:using Agela Venusil XBP C18 column(4.6 mm×250 mm,5μm),gradient elution with methanol-0.1%acetic acid as a mobile phase,the detection wavelength of 254 nm,and the flow rate of 1 mL·min^-1.The results show that the p-hydroxybenzoic acid in Momordica cochinchinensis is well separated with a separation degree of 1.75 and theoretical tray number of 126964.Moreover,the mass of p-hydroxybenzoic acid(x)shows good linear relationship with the peak area(y)within the range of 0.100-0.485μg,and the linear equation is y=1000000x-81274,(R^2=0.9997).Furthermore,the average adding standard recovery is 107.5%,and the content of p-hydroxybenzoic acid in Momordica cochinchinensis is 4.942μg·g^-1.The method is simple and efficient,which can be used for the determination of the content of p-hydroxybenzoic acid in Momordica cochinchinensis.
