摘 要:目的采用定量蛋白质组学串联质谱标签(tandem mass tag,TMT)技术筛选少精子症患者精子差异表达蛋白质,初步寻找少精子症的潜在生物学标志物,阐释相关的生物学过程及代谢通路,为少精子症的分子机制研究提供新的思路。方法随机选取少精子症患者和正常人精子样本,各30例。应用TMT技术寻找并鉴定差异表达蛋白质,对差异蛋白进行基因本体(gene ontology,GO)富集分析和KEGG通路分析。结果共鉴定1050种差异表达蛋白质,其中561种蛋白质上调表达,489种蛋白质下调表达。GO功能富集分析发现上述差异表达蛋白质参与了终止翻译和核转录mRNA分解代谢过程等生物过程;且主要分布在部分胞质和胞质核糖体中;分子功能是核糖体结构成分和结构分子活性等。KEGG信号通路分析涉及代谢通路、核糖体信号通路、氧化磷酸化信号通路、PI3K-Akt信号通路等。结论差异表达蛋白质的生物信息学有助于少精子症发病机制研究,可作为其分子机制研究的参考和补充。Objective To screen sperm differentially expressed proteins in patients with oligozoospermia by TMT technology,and to search for potential biomarkers of oligozoospermia preliminarily,to clarify related biological processes and metabolic pathways,and to provide new ideas for the molecular mechanism of oligozoospermia.MethodsThirty sperm samples from oligozoospermia and normal subjects were collected separately,and TMT technology was used to find and identify differentially expressed proteins.GO enrichment analysis and KEGG pathway analysis were performed on them.ResultsA total of 1050 differentially expressed proteins were identified,of which 561 were up-regulated and 489 were down-regulated.GO enrichment analysis showed that most of them were involved in biological processes such as termination of inversion and decomposition of nuclear transcription RNA,mainly distributed in some cytoplasmic and cytoplasmic ribosomes,and their molecular functions were ribosome structural components and structural molecules.KEGG signaling pathway analysis involves metabolic pathways,ribosomal signaling pathways,oxidative phosphorylation signaling pathways,and PI3K-Akt signaling pathways.ConclushionThe bioinformatics of differentially expressed proteins contributes to the pathogenesis of oligozoospermia and can be used as a reference and supplement for itsmolecular mechanism research.
