摘 要:通过熔融法以均苯四甲酸酐、尿素及相应的金属盐为原料,得到八羧基酞菁锰(MnOCPc)和八羧基酞菁铁(FeOCPc)。通过荧光光谱、紫外光谱、圆二色光谱等分析方法研究了金属酞菁与BSA的相互作用。结果表明,BSA的荧光发生了规律性的猝灭,其猝灭类型主要为静态猝灭。通过Stern-Volmer方程得到了两种金属酞菁对BSA的荧光猝灭常数和结合位点数等信息。圆二色光谱表明,金属酞菁使BSA的构象发生了变化,α-螺旋结构含量降低。抗氧化活性测试表明,金属酞菁、金属酞菁-BSA加合物均表现出一定的SOD活性,牛血清白蛋白的加入增强了金属酞菁的SOD活性。The FeOCPc(Fe-octacarboxyl ropathalocyanine)and MnOCPc(Mn-octacarboxyl pathalocyanine)were synthesized using pathalic anhydride,urea and ammoniam molybdate with melting method.The interaction between metal phthalocyanine and BSA was analyzed using fluorescence,UV,and circular dichroism.The results showed that metal phthalocyanines can cause the endogenous fluorescence quenching of BSA,and they are mainly static quenching.The information such as the quenching constant K SV,the binding constant K,and the binding method was calculated through Stern-Volme equation.Circular dichroism showed that the addition of metal phthalocyanine changes the conformation of BSA and decreases the content ofα-helical structure.The antioxidant activity tests of metal phthalocyanine and metal phthalocyanine-BSA adducts showed that FeOCPc and MnOCPc showed some SOD activities,and the addition of BSA can enhannce SOD activities of metal phthalocyanine.
