摘 要:该文以“解决问题”为导向,结合最新的科研成果,设计了一个化学生物学综合创新实验。实验依据荧光共振能量转移原理,将碳量子点和异硫氰酸荧光素进行偶联,制备了pH敏感的比率型荧光探针,并将该探针运用于胞内pH的动态监测。该实验体现了新型纳米材料合成、荧光光谱分析、细胞标记及细胞成像等前沿领域的相互交叉、相互融合。实验内容充实、结果稳定可重复。另外,实验涵盖了可供学生自主探索和实践的内容,有助于提升学生的实践创新能力。Based on the orientation of“problem solving”and the latest research results,a comprehensive innovative experiment of chemical biology is reported.According to the principle of fluorescence resonance energy transfer(FRET),a ratiometric pH sensor was prepared by covalently lingking carbon quantum dots(CDs)with fluorescein isothiocyanate(FITC),which was applied to monitor the intracellular pH in real time.The experiments show that the cross and integration of the front fields including the novel nanomaterials synthesis,fluorescence spectrometric analysis,cell labeling and imaging.This experiment reflects the intersection and fusion of cutting-edge fields such as the synthesis of new nanomaterials,fluorescence spectrum analysis,cell labeling,and cell imaging.The experimental content is substantial and the results are stable and repeatable.In addition,the experiment covers the content for students to explore and practice independently,which helps to improve the students’practical innovation ability.
