摘 要:目的探讨姜黄素改善大鼠肾缺血再灌注损伤的作用及其机制。方法夹闭肾动脉制后恢复供血制备肾缺血再灌注模型。用随机数字表法将大鼠分为3组:假手术组、模型组和实验组,每组15只。实验组用100 mg·kg^-1姜黄素预处理,假手术组和模型组用等量0.9%NaCl处理,共5 d。染色法观察大鼠肾组织变化并进行Paller评分,全自动生化分析仪检测大鼠肌酸酐(Cr)和尿素氮(BUN)含量,试剂盒法检测超氧化物歧化酶(SOD)和丙二醛(MDA)含量。结果假手术组、模型组和实验组Paller评分分别为(100.23±15.21),(602.30±28.36)和(375.32±18.32)分;这3组的BUN含量水平分别为(11.25±2.30),(26.89±5.65)和(20.32±1.98)mmol·L^-1;这3组的Cr含量水平分别为(40.32±1.23),(85.36±9.32)和(64.32±5.35)μmol·L^-1;这3组的SOD含量水平分别为(375.32±45.32),(250.21±25.32)和(312.21±12.32)U·mg^-1;这3组的MDA含量水平分别为(6.21±0.75),(10.02±1.23)和(7.01±0.77)mmol·mg^-1。上述指标:模型组与假手术组比较、或实验组与模型组比较,差异均有统计学意义(均P<0.01)。结论姜黄素可减轻缺血再灌注大鼠肾损伤程度,改善大鼠肾功能,这可能与姜黄素可以提高大鼠血清SOD含量、降低MDA含量有关。Objective To explore action mechanism of curcumin in improving renal ischemia-reperfusion(IR)in rats.Methods After pre-paration of clipping renal artery,blood supply was restored to prepare renal IR models.The random number table method was applied to divide them into three groups:sham operation group,model group and experimental group,with 15 rats in each group.The experimental group was given 100 mg·kg^-1 curcumin for pretreatment,while sham operation group and model group were treated with same amount of normal saline for 5 d.The changes of renal tissues were observed by staining and the a Paller score was carry out.The creatinine(Cr)and urea nitrogen(BUN)levels were determined by automatic biochemical analyzer.The contents of superoxide dismutase(SOD)and malondialdehyde(MDA)were determined by Kit.Results The Paller scores in sham operation group,model group and experimental group were(100.23±15.21),(602.30±28.36)and(375.32±18.32)point,respectively;the BUN level in the three groups were(11.25±2.30),(26.89±5.65)and(20.32±1.98)mmol·L^-1,respectively;the Cr level in the three groups were(40.32±1.23),(85.36±9.32)and(64.32±5.35)μmol·L^-1,respectively;the SOD content in the three groups were(375.32±45.32),(250.21±25.32)and(312.21±12.32)U·mg^-1,respectively;the MDA content in the three groups were(6.21±0.75),(10.02±1.23)and(7.01±0.77)mmol·mg^-1,respectively.Comparison between model group and sham operation group,or comparison between experimental group and model group,the difference of the factors were significantly(all P<0.01).Conclusion Curcumin can alleviate renal injury degree in IR rats,and improve renal function.The mechanism was related to increasing serum SOD content and decraeasing MDA content.
