摘 要:目的研究丙泊酚对结直肠癌细胞恶性表型的影响及其炎症机制。方法培养对数生长期人结肠癌细胞SW620,用0,10,50,100μmol·L^-1丙泊酚处理SW62048 h分别作为对照组和低、中、高3个浓度实验组。CCK-8实验检测细胞增殖能力,蛋白质印迹实验检测肿瘤坏死因子α(TNF-α)和细胞白细胞介素-1β(IL-1β)蛋白水平,实时荧光定量-PCR实验检测TNF-α和IL-1β的基因表达水平。结果对照组与低、中、高3个浓度实验组于72 h的细胞相对增殖能力(光密度值)分别为7.94±0.66,6.38±0.74,5.72±0.95和4.28±0.66;这4组的TNF-α蛋白相对表达水平(灰度值)分别为1.00±0.00,0.66±0.09,0.23±0.11和0.14±0.15;这4组的IL-1β蛋白相对表达水平(灰度值)分别为1.00±0.00,0.38±0.15,0.36±0.14和0.09±0.18。上述指标:低、中、高3个浓度实验组与对照组相比,差异均有统计学意义(P<0.05,P<0.01)。基因结果的趋势与蛋白一致。结论丙泊酚可抑制结直肠癌细胞的恶性增殖,同时下调IL-1β及TNF-α的表达,抑制炎症反应。Objective To investigate the effect of propofol on malignant phenotype of colorectal cancer cells and its inflammation mechanism.Methods Human colon cancer SW620 cells in logarithmic growth phase were cultured and treated with 0,10,50 and 100μmol·L^-1 propofol for 48 h as control group,experimental-L group,experimental-M group and experimental-H group,respectively.Cell Counting Kit-8 assay was used to detect cell proliferation.Western blot was used to detect tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)protein levels,and quantitative real-time-PCR was used to detect TNF-α and IL-1βmRNA expression levels.Results The relative proliferative ability(OD value)of cells in control group,experimental-L group,experimental-M group and experimental-H group at 72 h were 7.94±0.66,6.38±0.74,5.72±0.95 and 4.28±0.66,respectively;the relative protein expression levels(gray value)of TNF-α in the four groups were 1.00±0.00,0.66±0.09,0.23±0.11 and 0.14±0.15,respectively;the relative protein expression levels(gray value)of IL-1β in the four groups were 1.00±0.00,0.38±0.15,0.36±0.14 and 0.09±0.18,respectively.Comparison between three concentration experimental groups and control group,the difference of the factors were significantly(P<0.05,P<0.01).The trend of gene results is consistent with that of protein.Conclusion Propofol can inhibit the malignant proliferation of colorectal cancer cells,down-regulate the expression of IL-1β and TNF-α,and inhibit inflammation.
