摘 要:目的:观察炎性痛大鼠不同时间段前扣带回神经元放电活动情况及电针对其的干预作用。方法:将前扣带回微丝阵列电极植入成功的雄性SD大鼠完全随机分为空白组、模型组和电针组。模型组和电针组于足底注射完全弗氏佐剂建立大鼠炎性痛模型。动态观察患侧足跖机械痛阈和前扣带回神经元放电频率,计算各类神经元的变化率和分布。于造模后28d介入电针治疗,观察电针对炎性痛大鼠机械痛阈和神经元的变化率、分布的影响。结果:造模前各组大鼠基础机械痛阈无显著性差异;造模后各时间点,模型组大鼠机械痛阈显著低于空白组(P<0.01);电针治疗后可显著提升机械痛阈(P<0.01)。造模后1、14、28d,模型组大鼠前扣带回神经元放电频率变化率与空白组比较有显著性差异(P<0.01);造模后1、7、14、21d,模型组大鼠前扣带回抑制性、兴奋性和不变神经元构成比与空白组比较有显著性差异(P<0.05,P<0.01)。电针可显著改变炎性痛大鼠前扣带回抑制性、兴奋性和不变神经元构成比(P<0.01),对其放电频率变化率无影响。结论:炎性痛大鼠造模后各时间点前扣带回神经元放电频率、分布均有不同程度的变化;电针具有良好的镇痛作用,可能与改变前扣带回神经元放电分布相关。Objective:To observe neuronal electric activity of anterior cingulate cortex (ACC) at different time of inflammatory pain rats and intervention effect of electroacupuncture (EA) on it.Methods:Male SD rats which were implanted microwire array electrode successfully in ACC were completed randomly divided into normal group,model group and EA group.Through injecting subcutaneously with complete Freund’s adjuvant into right hind paw to establish inflammatory pain model of rats in model and EA group.Paw withdrawal threshold (PWT) and neuron spike frequency of ACC were measured before model and at 1d,7d,14d,21d and 28d after model.The change rate and distribution of all kinds of neurons were calculated by the firing frequency of neurons.EA stimulation on day 28 after model,we observed the effect of EA on PWT and neuron change rate and distribution.Results:There were no significantly different of the PWT among the normal group,model group and EA group.The PWT of model group was lower than that of normal group at every time point after model (P<0.01).Compared with model group,EA stimulation could increase the PWT (P<0.01).The change rate of neurons in the ACC of model group were significantly different with that of normal group on day 1,14,28 after model (P<0.01).The distribution of neurons in the ACC were also different with that of normal group on day 1,7,14,21 after model (P<0.05,P<0.01).EA could change the distribution of neurons in the ACC (P<0.01),but has no effect on the change rate of neurons.Conclusion:The change rate and distribution of all kinds of neurons in the ACC were different time points after model.EA has a good analgesic effect,which may be related to the change of the distribution of neurons in the ACC.
