摘  要:目的:探讨电针足三里和内庭对高脂血症SD大鼠脂肪组织腺苷酸活化蛋白激酶(AMPK)表达的影响。方法:33只SD雄性大鼠,随机分为高脂饮食组和正常饮食组,高脂血症造模成功后,高脂饮食组随机分为高脂电针组和高脂对照组,正常饮食组分为正常电针组和正常对照组,连续电针30d,腹主动脉采血法处死大鼠,RT-PCR法检测脂肪组织AMPKα2 mRNA表达,Western Blot法检测pAMPK蛋白表达。结果:造模后高脂饮食组血脂明显升高(P<0.05),高脂血症模型复制成功;电针30d后,高脂电针组血脂显著下降(P<0.05),针刺降脂有效。与高脂对照组比较,电针后高脂电针组脂肪组织pAMPKα蛋白表达量显著升高(P<0.05),脂肪组织AMPKα2 mRNA表达量无显著变化。结论:电针胃经穴位可以增加高脂血症大鼠脂肪组织AMPK蛋白表达。Objective:To investigate the effects of electroacupuncture (EA) on Zusanli (ST36) and Neiting (ST44) acupoint on AMPK expression in adipose tissue of SD rats with experimental hyperlipidemia.Methods:Thirty-three male SD rats were randomly divided into a high-fat diet group and a normal diet group.After successful modeling of hyperlipidemia,the high-fat diet group was randomly divided into a high-fat electroacupuncture group and a high-fat control group.Rats of blank EA group and hyperlipidemia EA group were electroacupunctured for 30 days.The rats were sacrificed with abdominal aorta blood sampling.RT-PCR method was used to detect the expression of AMPKα2 gene mRNA in adipose tissue,and the expression of AMPK protein was detected by Western Blot.Results:After modeling,blood lipids in the high-fat diet group were significantly increased (P<0.05),and the hyperlipidemia models were successfully copied.With treated by EA for 30d,the blood lipid of the high-fat electroacupuncture group decreased significantly (P<0.05),and acupuncture was effective in reducing lipids.The expression of pAMPKα protein in adipose tissue after electroacupuncture in the high-fat electroacupuncture group was significantly higher than that in the high-fat control group (P<0.05).No significant change in mRNA expression of AMPKα2 in adipose tissue after electroacupuncture in high-fat electroacupuncture group.Conclusion:EA at stomach meridian could increase AMPK protein expression in adipose tissue ofhyperlipidemia rats.
