摘 要:目的观察益心泰有效组分对心肌梗死后心力衰竭兔的心功能及心肌超微结构的影响。方法参照文献采用结扎左冠状动脉前降支法+PTU灌胃复制心肌梗死后心力衰竭兔模型,造模成功后,随机分为模型组,益心泰低、中、高剂量组及氯沙坦钾组。各组予以相应药物,连续灌胃4周后行心脏彩色多普勒超声检查,并采集心肌样本,行HE染色及透射电镜。结果与模型组相比,益心泰中、高剂量组与氯沙坦钾组心脏收缩功能指标(LVEF、LVFS)和舒张功能指标(E/A比值)升高(P<0.05),心脏结构性指标(LVIDs、LVIDd)降低(P<0.05),益心泰高剂量组与氯沙坦钾组心脏指数、左心室重量指数降低(P<0.05)。治疗效果上,益心泰高剂量组及氯沙坦钾组均优于益心泰低剂量组(P<0.05)。HE染色示:各治疗组的兔心肌细胞损伤程度均较模型组有不同程度的减轻,透射电镜镜下观察的结果也进一步验证HE染色的观察所见。结论益心泰有效组分可明显改善心肌梗死后心力衰竭兔的心功能及超微结构。Objective To observe the effect of Yixintai effective components on cardiac function and ultrastructure in rabbits with chronic heart failure after myocardial infarction. Methods Reference literature was used to help establish rabbit models of heart failure after myocardial infarction by ligation of the left anterior descending coronary artery + PTU. After the successful modeling, they were randomly divided into a model group, Yixintai low-, medium-and high-dose groups and a losartan potassium group. Each group was given the corresponding drug. After continuous gavage for 4 weeks, cardiac color Doppler ultrasonography was performed, and myocardial samples were collected for HE staining and transmission electron microscopy. Results Compared with the model group, the systolic function index(LVEF and LVFS) and diastolic function index(E/A ratio) increased in Yixintai medium-, high-dose group and the losartan potassium group(P< 0.05). The cardiac structural indicators(LVIDs, LVIDd) decreased(P < 0.05), and the heart index and left ventricular mass index decreased in the Yixintai high-dose group and losartan potassium group(P < 0.05). The therapeutic effect was better in the Yixintai high-dose group and the losartan potassium group than that in the low-dose group(P < 0.05). HE staining showed that the degree of myocardial cell damage in the rabbits in each treatment group was reduced to a certain extent as compared with that of the model group. Transmission electron microscopy also confirmed the observation of HE staining. Conclusion Yixintai effective components can significantly improve the cardiac function and ultrastructure in rabbits with heart failure after myocardial infarction.
