摘 要:目的研究沙棘多糖对巨噬细胞激活的影响以及对环磷酰胺诱导的免疫低下小鼠的免疫调节作用。方法体外培养巨噬细胞RAW264.7,采用CCK8法评价沙棘多糖对巨噬细胞体外增殖活力的影响;测定巨噬细胞上清液中一氧化氮(NO)的含量;采用酶联免疫吸附法检测细胞因子肿瘤坏死因子α(TNF-α)、白介素-6(IL-6)、γ-干扰素(IFN-γ)释放情况;同时用环磷酰胺建立免疫低下小鼠模型,分为对照组、模型组、沙棘多糖低剂量组以及高剂量组,分别测定小鼠的脾脏和胸腺指数、外周血白细胞计数、计算腹腔巨噬细胞吞噬百分率和吞噬指数,用聚合酶链式反应(PCR)法检测脾脏组织中TNF-α、IL-6、IFN-γmRNA的水平。结果在体外,沙棘多糖能够显著促进巨噬细胞的增殖,促进细胞因子TNF-α、IL-6、IFN-γ、NO的释放;在体内,沙棘多糖能够增强免疫低下小鼠的免疫器官指数、增加外周血白细胞数量、提升小鼠巨噬细胞的吞噬功能、提高脾脏组织中TNF-α、IL-6、IFN-γmRNA的水平。结论沙棘多糖能够激活巨噬细胞并且明显增强环磷酰胺致免疫低下小鼠的免疫功能。Objective To determine the immunomodulatory effect of hippophae rhamnoides polysaccharide on the macrophages and cyclophosphamide-induced immunocompromised mice. Methods RAW264.7 cells were cultured in vitro, and CCK8 method was used to evaluate the effect of hippophae rhamnoides polysaccharide on the proliferation of RAW264.7 cells. The content of NO in the supernatant of RAW264.7 cells was determined. The release of TNF-α, IL-6, and IFN-γ were detected by ELISA. In the in vivo experiments, mice were divided into a control group, a model group, a hippophae rhamnoides polysaccharide low dose group, and a hippophae rhamnoides polysaccharide high dose group. Cyclophosphamide was used to establish the immunosuppressed mouse model. The spleen and thymus index, peripheral blood leukocyte count, phagocytic percentage and phagocytic index of peritoneal macrophages were determined. The levels of TNF-α, IL-6, and IFN-γ mRNA in the spleen tissue were detected by PCR. Results In vitro, hippophae rhamnoides polysaccharide significantly promoted the proliferation of RAW264.7 cells, and promoted the release of TNF-α, IL-6, and IFN-γ, and NO of RAW264.7 cells. In vivo, hippophae rhamnoides polysaccharide significantly enhanced the immune organ index of the immunosuppressed mice, increased the number of peripheral blood leukocytes, enhanced the phagocytic function of macrophages, and improved the levels of TNF-α, IL-6, and IFN-γ mRNA in the spleen tissue. Conclusion hippophae rhamnoides polysaccharide can activate macrophages and enhance the immune function of cyclophosphamide-induced immunosuppressed mice.
