摘 要:目的研究加味酸枣仁汤水提物体内外主要化学成分。方法采用ACQUITY UPLC BEH C18色谱柱,以0.05%甲酸水-乙腈为流动相梯度洗脱,质谱扫描采用负离子模式检测,大鼠口服加味酸枣仁汤后,对加味酸枣仁汤的提取液、空白血浆和含药血浆进行分析,确定其体内外化学成分。结果通过与对照品对比、结合质谱数据和文献检索信息进行比较分析,鉴定了加味酸枣仁汤提取物中25个化学成分和大鼠血浆中的5个入血原形成分。结论酸枣仁皂苷A等5种化学成分确认为入血成分,本研究为加味酸枣仁汤中药效物质的代谢规律研究及其制剂开发中标志成分的选择提供了依据。Objective To analyze the main chemical constituents of modified Suanzaoren decoction(MSD) extracts in vitro and in vivo. Methods The separation was achieved on an ACQUITY UPLC BEH C18 column by gradient elution with mobile phase consisting of 0.05% formic acid in water(A) and acetonitrile(B). The ion acquisition was performed in a negative ion mode. After an oral administration of MSD, the blank plasma of rats, sample plasma of rats and extracts of MSD were analyzed to obtain the chemical constituents in vitro and in vivo of the formula. Results Based on the literature research, compared with standard substance and the data of MS/MS, 25 constituents were identified in MSD and 5 prototype components were characterized in the rat plasma. Conclusion Five chemical components including jujuboside A are confirmed as the components absorbed into blood. This research provides reference for further study of metabolism of the chemical constituents in MSD, and lays foundation for elucidating the primary components in pharmaceutical formulation.
