摘  要:目的建立高效液相色谱串联电喷雾检测器(HPLC-CAD)同时测定浓维磷糖浆中果糖、葡萄糖、蔗糖和麦芽糖含量的方法,为其质量标准提高提供依据。方法采用Alltech chrom Prevail carbohydrate ES 5u色谱柱(4.6 mm×250 mm,5μm),以乙腈-水(75∶25)为流动相,流速1.0 mL·min-1,柱温35℃。电喷雾检测器参数为:雾化温度35℃,采样频率10 Hz。结果上述4个糖类成分分离完全,线性关系良好(r均大于0.9990);精密度、重复性及回收率试验结果均符合含量测定要求,果糖、葡萄糖、蔗糖和麦芽糖的平均加样回收率分别为98.4%、97.5%、99.4%和95.4%。结论该方法灵敏度高、准确可靠,可用于浓维磷糖浆中果糖、葡萄糖、蔗糖和麦芽糖含量测定。Objective To establish a method for simultaneous determination of fructose,glucose,sucrose and maltose in Concentrated Divitamins and Sodium Phosphate Syrup by HPLC-CAD.Methods The analysis was performed on an Alltech chrom Prevail carbohydrate ES 5u column(4.6 mm×250 mm,5μm).The mobile phase was acetonitrile-water(75∶25)at a flow rate of 1 mL·min-1.The column temperature was controlled at 35℃.The detector of CAD was applied with nebulization temperature at 35℃,and collection frequency at 10 Hz.Results Under the above chromatographic conditions,the four saccharides was completely separated and showed good linear relationship with the correlation coefficients were not lower than 0.9990.The experiment results f precision,repeatability and recovery all accord with the requirements of content determination.The average recovery for fructose,glucose,sucrose and maltose were 98.4%,97.5%,99.4%and 95.4%respectively.Conclusion The method was high sensitivity,accuracy and reliability,and can be used for the determination of fructose,glucose,sucrose and maltose in Concentrated Divitamins and Sodium Phosphate Syrup.
