摘  要:目的探讨超声微泡靶向介导NT-3在治疗脊髓损伤(spinal cord injury,SCI)中的意义.方法40只SD大鼠分为4组.对照组打开椎板,未做打击;脊髓损伤组打击后不给任何处理;NT-3微泡组打击后经鼠尾静脉注射0.5 ml NT-3微泡;NT-3微泡+超声照射组打击后经鼠尾静脉注射0.5 ml NT-3微泡并进行超声照射.比较各组大鼠BBB(Basso Beattie Bresnahan)评分、脊髓组织中NSE、Caspase-3、IL-1β、IL-6和NT-3 mRNA的表达.结果与其他各组相比,NT-3微泡+超声照射组出现一定程度的BBB评分提高和细胞凋亡减少,而且残存神经元增加,炎症介质表达降低和NT-3 mRNA表达增加,差异均有统计学意义(P<0.05).结论超声微泡靶向介导NT-3对大鼠SCI有着积极的治疗作用,这对全面研究SCI的修复机制有着重要意义.Objective To explore the significance of ultrasonic microbubble-mediated NT-3 in the treatment of spinal cord injury.Methods Forty SD rats were divided into four groups.The vertebral lamina was opened in control group,but not given an impact.The rats in spinal cord injury(SCI)group were given an impact but no any treatment after impact.The rats in NT-3 microbubble group were injected with 0.5 ml NT-3 microbubbles via the tail vein of rats after SCI.The rats in ultrasonic microbubble-mediated NT-3 group were injected with 0.5 ml NT-3 microbubbles via the tail vein of rats after SCI and given ultrasound irradiation.The expression of NSE,Caspase-3,IL-1β,IL-6 and NT-3 mRNA in spinal cord was compared between these groups.Results Compared with the other groups,BBB score was improved in microbubble-mediated NT-3 group,the cell apoptosis was reduced,the residual neurons were increased,the inflammatory mediators were reduced and the expression of NT-3 mRNA was increased,and there was siginificant difference in the above parameters(P<0.05).Conclusion Ultrasonic microbubble-mediated NT-3 is beneficial to the repair of SCI in rats.It is of great significance to study the repair mechanism of SCI.
