摘  要:目的:观察电针对胰岛素抵抗大鼠海马胰岛素降解酶(IDE)、过氧化物酶体增殖物激活受体-γ(PPAR-γ)、β连环蛋白(β-catenin)与β淀粉样蛋白(Aβ42)表达的影响。方法:40只Wistar大鼠随机分为正常组、模型组、预防组和电针组,每组10只。电针治疗后,免疫组织化学法检测大鼠海马Aβ42阳性表达;实时荧光定量PCR检测大鼠海马IDE、PPARγ和β-catenin表达。结果:与正常组比较,其他各组大鼠海马Aβ42的阳性表达显著升高(P<0.01,P<0.05),IDE、PPARγ与β-catenin表达显著减少(P<0.01,P<0.05);与模型组比较,预防组、电针组大鼠海马Aβ42阳性表达显著减少(P<0.01),IDE、PPARγ和β-catenin表达显著增加(P<0.05,P<0.01);与预防组比较,电针组大鼠海马IDE、PPARγ表达显著减少(P<0.05)。结论:电针降低胰岛素抵抗大鼠海马Aβ42阳性表达,其作用机制可能与电针增加IDE、PPARγ、β-catenin表达相关。Objective:To observe the effects of electro-acupuncture on the expression of IDE,PPARγ,β-catenin and Aβ42 in hippocampus of insulin resistance rats.Methods:Forty Wistar rats were divided into 4 groups:normal group,model group,pre-EA group and EA group,10 rats each group.Expression levels of Aβ42 in hippocampus of IR rats;The expression of IDE,PPARγandβ-catenin mRNA in hippocampus were analyzed by real-time fluorescence quantitative PCR.Results:Compared with normal group,in other three groups,the expression levels of Aβ42 in hippocampus increased(P<0.01,P<0.05),the expression of IDE,PPARγandβ-catenin mRNA in hippocampus decreased(P<0.01,P<0.05)in other 3 groups;Compared with model group,in pre-EA group and EA group,the expression levels of Aβ42 in hippocampus decreased(P<0.01),the expression of IDE,PPARγandβ-catenin mRNA in hippocampus increased(P<0.05,P<0.01)in pre-EA group and EA group.Compared with pre-EA group,the expression of IDE,PPARγmRNA in hippocampus decreased(P<0.05)in EA group.Conclusion:The mechanism of electro-acupuncture improvement on expression levels of Aβ42 in insulin resistance model rats,is pertaining to the regulation of regulating the expression of IDE,PPARγandβ-catenin mRNA in hippocampus.
