摘 要:目的:探讨回阳生肌膏影响糖尿病大鼠阴证疮面的微血管新生可能的作用机制。方法:108只大鼠剔除实验术中死亡的18只大鼠后,将剩余90只大鼠随机分为:实验组、实验+抑制剂组、模型+抑制剂组、模型组、对照组,每组18只,实验组和实验+抑制剂组给予回阳生肌膏纱条,其余各组给予凡士林纱条,实验+抑制剂组和模型+抑制剂组腹腔注射Ly294002抑制剂,其余各组腹腔注射0.9%氯化钠溶液。每天给药1次,分别于给药后3、7、10d每组各处死6只大鼠,观察CD34免疫组化染色情况,ELISA法检测VEGF的含量变化。结果:给药3、7、10d,实验组较模型组CD34的IOD值升高显著(P<0.01);给药7d,实验+抑制剂组较实验组CD34的IOD值显著下降(P<0.01),模型+抑制剂组较模型组CD34的IOD值显著下降(P<0.01);给药3、7、10d实验组和实验+抑制剂组较模型组VEGF的蛋白含量显著升高(P<0.01),模型+抑制剂组较模型组VEGF的蛋白含量显著降低(P<0.01)。结论:回阳生肌膏可能通过激活PI3K/AKT通路,促进血管新生,从而促进伤口愈合。Objective:To explore the possible mechanism of Huiyang Shengji Ointment affecting the microvascular neogenesis of yin syndrome wound of diabetes rats.Methods:A total of 108 rats were randomly divided into the experimental group,the experiment+inhibitor group,the model+inhibitor group,the model group,the control group,18 rats in each group.The experimental group and the experimental+inhibitor group were given Huiyang Shengji Ointment,the other groups were given Vaseline gauze,the experimental+inhibitor group and the model+inhibitor group were intraperitoneally injected with Ly294002 inhibitor,and the other groups were intraperitoneally injected with normal saline.The rats were administered once a day,and 6 rats were sacrificed in each group 3 days,7 days,and 10 days after each administration.The immunohistochemical staining of CD34 was observed.ELISA was used to detect the change of VEGF content.Results:After 3,7,10 days of administration,the IOD value of the CD34 in the experimental group was significantly higher than that in the model group(P<0.01);After 7 days,the IOD value of the CD34 in the experimental+inhibitor group was significantly lower than that in the experimental group(P<0.01).Compared with the model group,the IOD value of CD34 in the model+inhibitor group lowered significantly(P<0.01);the protein content of VEGF in the experimental group and the experimental+inhibitor group was significantly higher than that of the model group at 3,7,10 days(P<0.01),the protein content of VEGF in the model+inhibitor group was significantly lower than that in the model group(P<0.01).Conclusion:Huiyang Shengji Ointment may promote microvascular neogenesis of diabetes yin syndrome wound by activating the PI3K/AKT pathway.
