摘  要:目的:探索黄芪、水蛭有效组分对经脂多糖(LPS)诱导增生的大鼠肾小球系膜细胞(GMC)核转录因子抑制因子(IκB)、核转录因子(NF-κB)、血小板源性生长因子-BB(PDGF-BB)表达的影响。方法:LPS诱导GMC增殖后,分别采用黄芪多糖、黄芪甲苷、毛蕊异黄酮、水蛭素进行干预,24h后收集细胞,RT-PCR法测定PDGF-BB mRNA的表达水平,Western Blot法测NF-κB、IκB蛋白的表达。结果:LPS诱导GMC增殖后,黄芪多糖、黄芪甲苷、毛蕊异黄酮及水蛭素能够降低PDGF-BB mRNA和NF-κB蛋白的表达(P<0.05),而黄芪多糖、黄芪甲苷、毛蕊异黄酮均可升高IκB蛋白的表达(P<0.05)。结论:黄芪、水蛭有效组分能够通过影响PDGF-BB、IκB、NF-κB的表达减少GMC的增殖及细胞外基质的沉积,延缓肾小球硬化的进展。Objective:To explore the effects of effective components in milkvetch root and leeches on the lipopolysaccharides(LPS)-induced nuclear transcription growth factor inhibitors(IκB),nuclear transcription growth factor(NF-κB)and platelet-derived growth factor-BB(PDGF-BB)expression of glomerular mesangial cells(GMCs)in rats.Methods:LPS induced rat GMCs proliferation,respectively using milkvetch root polysaccharides,milkvetch root glycosides,calycosin,hirudin intervention,collecting cells after 24 hours,RT PCR method for determining the expression of PDGF-BB mRNA level changes,Western Blot method measuring the NF-κB,IκB protein expression of predominate.Results:After LPS induced GMCs proliferation,milkvetch root polysaccharides,milkvetch root glycosides,calycosin,hirudin could reduce the expression of NF-κB protein and PDGF-BB mRNA(P<0.05),while milkvetch root polysaccharides,milkvetch root glycosides,and calycosin could increase the expression of IκB protein(P<0.05).Conclusion:The effective components of milkvetch root and leech can reduce the proliferation of mesangial cells and the deposition of extracellular matrix by affecting the expression of PDGF-BB,IκB and NF-κB,thus delaying the progression of glomerulosclerosis.
