摘 要:目的:探讨吴茱萸汤对芥子油内脏痛模型小鼠止痛作用机制。方法:45只C57BL/6雄性小鼠随机分为对照组,模型组,吴茱萸汤低、中、高剂量组,每组9只。吴茱萸汤低、中、高剂量组分别灌以吴茱萸汤水煎液3.12、6.24、12.48g·kg^-1·d^-1,共7d,第7天,吴茱萸汤各剂量组灌胃后2h,0.75%芥子油(50μL/只)灌肠诱导小鼠内脏痛模型,观察吴茱萸汤干预后的疼痛行为变化,对小鼠结肠组织进行免疫组化染色及HE染色,Western Blot检测吴茱萸汤干预后的小鼠结肠组织中瞬时感受器电位锚蛋白-1(TRPA1)和瞬时感受器电位香草素受体1(TRPV1)的表达变化。结果:HE染色可见,吴茱萸汤各剂量预处理7d后对结肠黏膜具有不同程度的保护作用。行为学实验中,不同浓度吴茱萸汤预处理后,小鼠疼痛行为频次和疼痛行为出现时间呈现不同程度降低和延迟(P<0.01)。免疫组化和Western Blot结果显示,吴茱萸汤干预后小鼠结肠中的TRPA1、TRPV1表达均显著下调(P<0.01)。结论:热敏通道TRPA1、TRPV1是吴茱萸汤发挥止痛作用,尤其是缓解芥子油内脏痛的关键靶点。Objective:To investigate the analgesic effect of Wuzhuyu Decoction(WZYT)on mustard oil(MO)-induced visceral pain in mice.Methods:A total of 45 C57BL/6 male mice were randomly divided into control group,model group,low-dose WZYT group(3.12g·kg^-1·d^-1),mid-dose WZYT group(6.24g·kg^-1·d^-1)and high-dose WZYT group(12.48g·kg-1·d^-1)were administered respectively to three groups by gavage for 7 days.On the 7th day,2 hours after the administration of WZYT,0.75%MO(50μL per mouse)was used to induce acute visceral pain mode through enema.Animal behavior experiment,immunohistochemistry technique,HE stain and Western Blot were used to observe the analgesic effect of WZYT and the expression of TRPA1 and TRPV1 in colon tissue of MO-induced visceral pain mice.Results:HE showed that after 7 days of pretreatment with different doses of WZYT,it has different protective effects on colonic mucosa.In the behavioral experiment,after pretreatment with different concentrations of WZYT,the number of behaviors and the latency of first behavior in mice showed different degrees of reduction and delay(P<0.01).Immunohistochemistry and Western Blot showed that the expressions of TRPA1 and TRPV1 in the colon of mice were significantly down-regulated after the intervention of WZYT(P<0.01).Conclusion:Thermosensory channels TRPA1 and TRPV1 play a key role on the analgesic effect of WZYT.
