新诊断2型糖尿病患者中医辨证分型与TCF7L2基因单核苷酸多态性的相关性

摘　　要：目的:探讨新诊断2型糖尿病患者的中医辨证分型与2型糖尿病易感基因TC F7L2单核苷酸多态性(SNPs)的相关性。方法:选择174例新发2型糖尿病患者,中医辨证分型分别为痰热互结型、热盛伤津型、气阴两虚型,另选择健康人50名作为正常对照。检测临床生化并记录年龄、性别、血压、身高、体质量、腰围、臀围,计算体重指数。利用PCR扩增测序技术将TCF7L2基因rs7903146位点、rs12255372位点SNPs进行分析。结果:TCF7L2基因rs7903146位点SNPs变异型T等位基因在各组中分布差异有统计学意义(P<0.05),痰热互结组显著高于正常对照组和热盛伤津组(P<0.05)。TCF7L2基因rs12255372位点SNPs变异型T等位基因在各组中分布频率差异无统计学意义。结论:TCF7L2基因rs7903146位点SNPs变异型T等位基因可能和新诊断2型糖尿病痰热互结型有相关性。Objective:To investigate the relationship between TCM syndrome differentiation of newly diagnosed T2DM and single nucleotide polymorphism of T2DM susceptibility gene TCF7L2.Methods:A total of 174 patients with newly diagnosed T2DM were enrolled.TCM syndrome differentiation was divided into phlegm-heat interaction type,overheat damage fluid type,deficiency of both qi and yin type.Another 50 healthy people were selected as normal controls.Test clinical biochemistry indicators.Record the age,sex,blood pressure,height,weight,waist circumference,hip circumference,and calculate the body mass index.The single nucleotide polymorphisms of the rs7903146 and rs12255372 loci of the TCF7L2 gene were classified by PCR amplification and sequencing.Results:There was a statistical difference in the distribution of single nucleotide polymorphisms variant T alleles at rs7903146 locus of TCF7L2 gene in each group(P<0.05),the distribution in phlegm-heat interaction type group was significantly higher than and normal control group and overheat damage fluid type group(P<0.05).There was no statistical difference in the distribution of T alleles at rs12255372 locus in each group.Conclusion:The single nucleotide polymorphism T alleles of the TCF7L2 gene at rs7903146 locus may be associated with phlegm-heat interaction type in newly-diagnosed T2DM patients.
