醒脑解郁胶囊对卒中后抑郁模型大鼠外周血T淋巴细胞亚群的影响

摘　　要：目的:观察醒脑解郁胶囊对卒中后抑郁(PSD)模型大鼠外周血T淋巴细胞亚群CD3^+、CD4^+、CD8^+的影响。方法:采用大脑中动脉闭塞联合孤养法与皮下注射利血平制备PSD大鼠模型,中药早期干预组与西药早期干预组造模14d,同时分别给予中药、西药连续灌胃35d。中药治疗组与西药治疗组造模14d,再给予中药、西药灌胃21d。体质量测量及糖水消耗实验、旷野实验等行为学评估大鼠抑郁状态,流式细胞术检测大鼠外周血CD3^+、CD4^+、CD8^+的含量。结果:与空白组比较,PSD造模成功后的各组大鼠体质量及糖水消耗量显著下降(P<0.05,P<0.01),旷野实验所测的运动得分显著降低(P<0.01,P<0.05);给药治疗后,与模型组比较,中药早期干预组、西药早期干预组、中药治疗组及西药治疗组上述指标均显著升高(P<0.01,P<0.05),且CD4^+含量显著增加(P<0.01);中药早期干预组与西药早期干预组CD8^+含量显著增加(P<0.05,P<0.01)。结论:醒脑解郁胶囊可改善模型大鼠的体质量,减轻症状及行为学状态,对PSD模型大鼠外周血CD3^+及CD4^+/CD8^+比例无明显影响,但可增加CD4^+与CD8^+的含量。Objective:To observe the effect of Xingnao Jieyu Capsules on CD3^+,CD4^+and CD8^+in the peripheral blood T lymphocyte subsets in post-stroke depression(PSD)model rats.Methods:PSD model rats were prepared by using middle cerebral artery occlusion combined with solitary curing and subcutaneous injection of reserpine.The early intervention group of traditional Chinese medicine(TCM)and the early intervention group of western medicine were modeled for 14 days,while the traditional Chinese medicine and western medicine were given continuous gavage for 35 days respectively.The TCM treatment group and the western medicine treatment group were modeled for 14 days,followed by the TCM or western medicine gavage for another 21 days.The depression status of rats were evaluated by weight measurement,sugar water consumption test,open field test,and the contents of CD3^+,CD4^+and CD8^+in the peripheral blood of rats were detected by flow cytometry.Results:Compared with the blank group,the weight and sugar water consumption of the rats in each group were significantly decreased(P<0.05,P<0.01),and the exercise scores measured in the field experiment were significantly decreased(P<0.01,P<0.05).Compared with the model group,the early intervention group of TCM,the early intervention group of western medicine,the treatment group of TCM and the treatment group of western medicine were significantly increased(P<0.01,P<0.05),and the content of CD4^+was significantly increased(P<0.01),The CD8^+content of the early intervention group and the western medicine early intervention group increased(P<0.05,P<0.01).Conclusion:Xingnao Jieyu Capsules can improve the weight loss symptoms and behavioral status of the model rats.it has no significant on peripheral blood CD3^+and the rate of CD4^+/CD8^+in PSD model rats,however,it can increase the content of CD4^+and C8^+.
