摘  要:目的探讨五味子乙素(Schizandrin B,Sch B)对慢性酒精中毒大鼠学习记忆能力的作用及对大鼠海马组织PSD-95蛋白表达的影响。方法40只成年Wistar雄性大鼠随机分为正常组、模型组、Sch B低剂量组、Sch B中剂量组和Sch B高剂量组。除正常组给予蒸馏水灌胃,其余4组建立慢性酒精中毒动物模型,56°北京红星二锅头白酒连续灌胃8周;动物模型建立成功后,Sch B低、中、高剂量组分别按10 mg/kg、20 mg/kg、40 mg/kg给予Sch B灌胃,1 d 1次,连续灌胃30 d。应用Morris水迷宫检测各组大鼠的学习记忆能力;采用Western Blot免疫印迹法检测各组大鼠大脑海马组织中的突触后致密蛋白95(postsynaptic density protein-95,PSD-95)蛋白的表达。结果Morris水迷宫实验结果显示,与正常组比,模型组潜伏期明显变长,而平台穿越次数变少(P<0.05);与模型组比,Sch B低、中剂量组潜伏期逐渐缩短,而平台穿越次数逐渐增多,高剂量组潜伏期明显缩短(P<0.05),且平台穿越次数明显增多(P<0.05);Western Blot印迹法实验结果显示,与正常组比较,模型组大鼠海马组织中的PSD-95蛋白表达减少(P<0.05);与模型组相比,Sch B低、中、高剂量组海马组织中的PSD-95蛋白表达逐渐增高,中、高剂量组PSD-95蛋白表达水平明显增高(P<0.05)。结论慢性酒精中毒可引起大鼠学习记忆能力障碍,Sch B能明显改善慢性酒精中毒大鼠学习记忆能力障碍,其可能通过调控大鼠大脑海马组织中PSD-95蛋白的表达水平实现。Objective To investigate the effects of Schizandrin B(Sch B)on learning and memory impairment in rats with chronic alcoholism and the effect on the expression of postsynaptic density protein-95(PSD-95)protein in hippocampus tissues of rats.Methods Forty male adult Wistar rats were randomly divided into a normal group,a model group,a low-dose Sch B group,a medium-dose Sch B group and a high-dose Sch B group.In addition to the normal group which was given intragastrical gavage of distilled water,the other four groups were established into animal models with chronic alcoholism.The rats were continuously given intragastrical gavage of 56°Beijing Red Star Erguotou liquor for 8 weeks.After the animal models were developed successfully,the low-dose,medium-dose and high-dose Sch B groups were respectively given intragastric gavage of Sch B at doses of 10 mg/kg,20 mg/kg and 40 mg/kg,once daily for 30 days.Morris water maze was used to detect the learning and memory ability of rats in every group.Western Blot was used to detect the expression of PSD-95 protein in the brain hippocampus tissues of rats in each group.Results Morris water maze test results showed that compared with the normal group,the model group had obviously lengthened latency but reduction of crossing platform times(P<0.05);and compared with the model group,the low-and medium-dose Sch B groups had gradually shortened latency but grdual increase of crossing platform times,and high-dose Sch B groups had obvious shortened latency(P<0.05),moreover,obvious increase of crossing platform times(P<0.05).Western Blot results showed that compared with normal group,the expression of PSD-95 protein in hippocampus tissues of rats in the model group was reduced(P<0.05);and compared with the model group,the expressions of PSD-95 protein in hippocampus tissues of rats in the low-dose,medium-dose and high-dose Sch B groups were gradually increased,and those in the medium-dose and high-dose Sch B groups were significantly increased(P<0.05).Conclusion Chronic alcoholism can
