摘  要:目的观察菟丝子黄酮及槲皮素对卵巢早衰(POF)大鼠性激素水平及卵巢卵泡的影响。方法将动情周期正常的雌性大鼠按体重随机分为5组:正常组、模型组、雌二醇组、菟丝子黄酮组和槲皮素组,每组各10只。除正常组以外,其余各组大鼠灌胃雷公藤多苷75 mg·kg^-1·d^-1制备卵巢早衰模型的同时,雌二醇组灌胃雌二醇0.1 mg·kg^-1·d^-1,槲皮素组灌胃槲皮素600 mg·kg^-1·d^-1,菟丝子黄酮组灌胃菟丝子黄酮530.1 mg·kg^-1·d^-1,均连续30 d。计算卵巢指数,以酶联免疫吸附法法检测抗苗勒氏管激素(AMH)、雌二醇(E2)、卵泡刺激素(FSH)和黄体生成素(LH)水平。结果模型组、雌二醇组、槲皮素组和菟丝子黄酮组的卵巢指数分别为(0.32±0.03),(0.38±0.05),(0.39±0.06)和(0.39±0.06)mg·g^-1;这4组的E2分别为(136.54±14.42),(201.21±15.19),(193.48±11.73)和(189.60±13.84)mU·mL^-1;这4组的LH分别为(1.41±0.22),(1.28±0.03),(1.29±0.10)和(1.34±0.08)mU·mL^-1。上述指标:雌二醇组、槲皮素组和菟丝子黄酮组与模型组比较,差异均有统计学意义(P<0.05或P<0.01)。结论菟丝子黄酮和槲皮素能在一定程度上改善雷公藤多苷致POF模型的卵巢功能,槲皮素可能是菟丝子黄酮中治疗POF的主要有效成分。Objective To observe the effects of Cuscuta flavonoids and quercetin on the levels of sex hormones and ovarian follicles in rats with premature ovarian failure(POF).Methods Female rats with normal estrous cycle were randomly divided into 5 groups according by weight:normal group,model group,estradiol group,dodder flavonoid group and quercetin group,with 10 rats in each group.Except for the normal group,rats in the other groups were intragastrically administered with 75 mg·kg^-1·d^-1 tripterygium wilfordii to during the POF model,the estradiol group was gavage 0.1 mg·kg^-1·d^-1 estradiol,quercetin group gavage 600 mg·kg^-1·d^-1 quercetin,Cuscuta flavonoid group gavage 530.1 mg·kg^-1·d^-1 Cuscuta flavonoids for 30 consecutive days.Ovarian index was calculated.Anti-mullerian hormone(AMH),estradiol(E2),follicle-stimulating hormone(FSH),luteinizing hormone(LH)were detected by enzyme linked immunosorbent assay.Results The ovarian index in model group,estradiol group,quercetin group,and Cuscuta flavonoid group were(0.32±0.03),(0.38±0.05),(0.39±0.06)and(0.39±0.06)mg·g^-1;E2 in the four groups were(136.54±14.42),(201.21±15.19),(193.48±11.73),(189.60±13.84)m U·m L^-1;LH in the four groups were(1.41±0.22),(1.28±0.03),(1.29±0.10)and(1.34±0.08)m U·m L^-1.Comparison between estradiol group,dodder flavonoid group and quercetin group with model group,the difference of the factors were significantly(P<0.05 or P<0.01).Conclusion Cuscuta flavonoids and quercetin can improve the ovarian function of the POF model induced by tripterygium glycosides to some extent.Quercetin may be the main active ingredient in the treatment of POF in Cuscuta flavonoids.
