摘 要:目的探索瓜子金总皂苷的最佳提取工艺。方法选取提取时间、液料比和乙醇浓度为考察因素,基于提取方式预选及单因素实验检测结果,应用Box-Behnken中心组合建立数学模型,以总皂苷提取率为响应值进行响应面分析,从而得到最佳工艺。结果通过Box-Behnken优化设计的最佳提取方案为乙醇浓度73%、液料比89 ml/g、微波提取时间53 min;总皂苷提取率预测值为8.96%,实测值为9.07%,相对误差为1.21%。结论本研究所得瓜子金总皂苷提取工艺科学可靠,可为瓜子金药材的开发奠定初步基础。Objective To explore the best extraction technology of total saponins from Polygala japonica Houtt.Methods Extraction time,liquid-solid ratio and ethanol concentration were selected as influencing factors during extraction.Based on preselection extraction method experiment and single-factor tests results to establish mathematical model,by Box-Behnken design.The total saponins extraction rate was detected and used as an response surface index,then evaluate and get the best extraction technology.Results The optimum conditions for total saponins were extraction time 53 min,ethanol concentration 73%,liquid-solid ratio 89 ml/g.The predicted extraction yield of total saponins was 8.96%,and the verification 9.07%,with relative error of 1.21%.Conclusion The optimized extraction technology,which lay the initial foundation for the development of Polygala japonica Houtt is scientific and reliable.
