摘 要:目的:观察消渴平合剂对糖尿病肾病db/db小鼠肾脏足细胞裂孔隔膜蛋白Nephrin、Podocin、CD2AP表达的影响,探讨其防治糖尿病肾病的机制。方法:选取12只db/m小鼠为正常对照组,其余36只db/db小鼠随机分为模型组、消渴平组、厄贝沙坦组,每组各12只;连续给药8周后分别检测各组小鼠空腹血糖(FBG)、胱抑素C(CysC)及24 h尿蛋白(24 hUP)。采用Real-time PCR、Western blot检测肾脏足细胞nephrin、podocin及CD2AP mRNA和蛋白表达变化。结果:消渴平合剂可以明显降低db/db小鼠FBG、CysC和24 hUP。与正常对照组相比,模型组小鼠肾脏足细胞nephrin、podocin及CD2AP的表达明显降低(P<0.01);与模型组相比,消渴平组nephrin、podocin及CD2AP的表达明显上调(P<0.05),与厄贝沙坦组比较无明显差异(P>0.05)。结论:消渴平合剂可以降低db/db小鼠空腹血糖、胱抑素C及24 h尿蛋白,保护肾功能,其机制可能与上调肾脏足细胞nephrin、podocin和CD2AP表达有关。Objective:To observe the effects of Xiaokeping Mixture on the expressions of nephrin,podocin and CD2AP in renal podocytes of db/db mice,and to explore the mechanism of its prevention and treatment of diabetic kidney disease.Methods:12 db/m mice were selected as the normal control group,and another 36 db/db mice were randomly divided into model group,Xiaokeping group,and Irbesartan positive group,with 12 mice in each group.After 8 weeks of continuous administration,fasting blood glucose(FBG),cystatin C(CysC),and urine protein of 24 hours(24 hUP)in each group were measured,Expressions of nephrin,podocin and CD2AP in renal podocytes were detected by Western blot and real-time PCR.Results:Xiaokeping Mixture could significantly reduce fasting blood glucose,cystatin C and 24 h urinary protein in db/db mice.Compared with the normal control group,nephrin,podocin and CD2AP expressions of renal podocytes in the model group were significantly decreased(P<0.01);Compared with the model group,the expressions of nephrin,podocin and CD2AP were significantly up-regulated in the Xiaokeping group(P<0.05),and there was no significant difference was found between the Xiaokeping group and Irbesartan group(P>0.05).Conclusion:Xiaokeping Mixture can reduce fasting blood glucose,cystatin C and 24 h urinary protein in db/db mice,so it may protect renal function.The mechanism may be related to the up-regulation of nephrin,podocin and CD2AP expression of renal podocin cells.
