摘  要:目的:探讨姜黄素对动脉粥样硬化(AS)家兔的保护作用及可能的作用机制。方法:72只雄性新西兰家兔随机分为假手术组、模型组、阳性药物组和姜黄素组,每组l8只。高脂饲料喂养、液氮冷冻造模,姜黄素组给予姜黄素200 mg/(kg·d)灌胃,阳性药物组予瑞舒伐他汀2.5 mg/(kg·d)灌胃,模型组和假手术组均给予相同剂量的生理盐水灌胃。分别于术后第4、8、12周检测血糖(BG)、三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)、超敏C-反应蛋白(hs-CRP)、肿瘤坏死因子(TNF-α)、白介素-6(IL-6)、超氧化物歧化酶(SOD)、一氧化氮(NO)、内皮素(ET)等指标。结果:第8周时,姜黄素组和阳性药物组血脂较模型对照组降低,差异有统计学意义(P<0.05),在第12周时差异更明显(P<0.01)。模型组较假手术组炎症指标hs-CRP、TNF-α、IL-6增高,差异有显著统计学意义(P<0.01);姜黄素组和阳性药物组炎性指标较模型组均降低(P<0.05或P<0.01)。模型组较假手术组SOD活性明显降低,而姜黄素组较模型组SOD活性增强,差异有统计学意义(P<0.01),阳性组与模型组比较差异无统计学意义(P>0.05)。模型组较假手术组NO水平降低而ET水平升高(P<0.01),姜黄素组和阳性药物组NO水平逐渐回升而ET水平降低,与模型组比较差异有统计学意义(P<0.01)。结论:姜黄素对AS有干预作用,其机制与降脂、降低炎症反应、抗氧化和保护修复内皮功能相关。Objective:To investigate the protective effect of Curcumin on atherosclerosis(AS)in rabbits and its possible mechanism.Methods:72 male New Zealand rabbits were randomly divided into sham operation group,model group,positive drug group and Curcumin group,with l8 rabbits in each group.The Curcumin group was given Curcumin 200 mg/(kg·d)by gavage,the positive drug group was given rosuvastatin 2.5 mg/(kg·d)by gavage,and the model group and the sham group were given the same dose of normal saline.Blood glucose(BG),three acyl glycerin(TG),total cholesterol(TC),low density lipoprotein(LDL),allergic-C reaction protein(hs-CRP),tumor necrosis factor(TNF alpha),interleukin 6(IL-6),superoxide dismutase(SOD),nitric oxide(NO),endothelin(ET),and other indicators were detected respectively at 4,8,12 weeks.Results:At week 8,serum lipids in the Curcumin group and the positive drug group were lower than those in the model control group,with statistically significant differences(P<0.05),and more significant differences at week 12(P<0.01).hs-CRP,TNF-α,IL-6 were significantly higher in the model group than in the sham group(P<0.01).The inflammatory indexes of Curcumin group and positive drug group were lower than those of model group(P<0.05 or P<0.01).SOD activity in the model group was significantly lower than that in the sham group,while SOD activity in the Curcumin group was significantly higher than that in the model group(P<0.01).There was no significant difference between the positive group and the model group(P>0.05).Compared with the sham group,the NO level in the model group decreased while the ET level increased(P<0.01).The NO level in the curcumin group and the positive drug group gradually increased while the ET level decreased,showing a statistically significant difference compared with the model group(P<0.01).Conclusion:Curcumin has an interventional effect on AS,and its mechanism is related to lowering lipids,reducing inflammation,anti-oxidation and protecting and repairing endothelial function.
