摘  要:目的:探讨不同浓度的百草枯(PQ)诱导人胚肺成纤维细胞纤维化(MRC?5)构建细胞纤维化模型。方法:运用CCK?8(cellcountingkit?8)法检测不同浓度的PQ对MRC?5细胞的毒性,以测得的最高不致死给药浓度的PQ诱导的MRC?5细胞模型,进而通过实时荧光定量PCR(qRT?PCR)检测细胞纤维化指标的mRNA及蛋白质表达水平变化。结果:50μg/ml PQ可成功诱导MRC?5细胞发生纤维化,且电镜下细胞成纤维化及细胞内α?平滑肌肌动蛋白的mRNA表达水平均显著升高。结论:PQ诱导MRC?5细胞纤维化的表型,为后续实验研究提供基础。Objective:To investigate the different concentrations of paraquat(PQ)induced fibrosis in human embryonic lung fibroblasts(MRC?5)to construct a cellular fibrosis model.Methods:CCK?8(cellcountingkit?8)was used to detect the toxicity of different concentrations of PQ to MRC?5 cells.PQ?induced mrc?5 cell model with the highest measured nonlethal drug delivery concentration,and then the mrna and protein expression levels of cell fibrosis indexes were detected by real?time fluorescence quantitative PCR(qRT?PCR).Results:The results showed that 100μg/ml,PQ could successfully induce fibrosis in MRC?5 cells,and the mRNA expression levels of intracellularα?smooth muscle actin were significantly increased.Conclusion:The phenotype of PQ?induced fibrosis in MRC?5 cells provides the basis for subsequent experimental studies.
