摘 要:以大葱未开放花蕾为试材,研究不同基因型和低温、热激两种预处理方式对大葱离体雌核发育诱导单倍体效果的影响。结果表明:不同基因型大葱以鲁葱杂5号的胚状体诱导率最高,为4.50%,再生植株数为15株;低温处理的胚状体诱导率和再生植株数整体高于热激处理,两指标分别于低温处理3 d、热激处理36 h时达到最大值,分别为7.50%、41株和3.50%、17株;基因型和预处理试验分别得到31、147株再生植株,其中,分别有4、18株被鉴定为单倍体,单倍体诱导率分别为12.90%和12.24%。The haploid induction capacity of spring onion by gynogenesis in vitro was studied with buds of different genotypes under low temperature and heat shock pretreatment conditions.The results showed that fifteen regenerated plants were cultured from Lucongza 5 with the highest embryogenesis induction rate of4.50%.Compared with heat shock treatment,the low temperature treatment resulted in higher embryogenesis induction rate and more regenerated plants,which reached the maximum as 7.5%and 41 plants under low temperature for 3 days and 3.5%and 17 plants under heat shock treatment for 36 hours,respectively.In the genotype and pretreatment experiments,31 and 147 spring onion regenerated plants were obtained,in which,four and 18 regenerated plants were identified as haploid,respectively.The haploid induction rates were12.90%and 12.24%respectively.
