摘 要:独一味(Lamiophlomis rotata)是青藏高原濒危保护药用植物。为研究外源钙对野生独一味抗逆性的影响,在玛曲县独一味野生抚育基地分别选取36个成株样方及36个幼苗样方,在越冬前不同时期对幼苗及成株叶面喷施不同浓度的CaCl2(0、5、10、15、20\,25 mmol·L-1),次年采集各处理样品带回实验室测定独一味叶片膜稳态性、渗透调节机能、膜质过氧化和抗氧化酶活性及根系TTC还原活力等抗逆指标,旨在探寻改善其越冬返青生理特性的处理方法,为独一味野生抚育和驯化栽培提供科学依据。结果表明:外源钙处理浓度和喷施次数对越冬后独一味抗逆指标具有显著影响,以喷施2次15 mmol·L-1 CaCl2的作用效果最佳,通过调控叶片可溶性糖和抗氧化酶活性保护膜系统的稳态性和完整性,使叶片相对电导率和丙二醛(MDA)含量维持在较低的水平,增强了根系活力,有效提高了野生独一味的返青率,在独一味野生抚育中具有潜在的应用价值。Lamiophlomis rotata is an endangered medicinal plant distributed in the Qinghai-Tibet Plateau.In order to study the effects of exogenous Ca2+on the L.rotata,36 plants and 36 seedlings were selected at Maqu Wild Tending Base,different concentrations of CaCl2(0,5,10,15,20 and 25 mmol·L-1)were sprayed at different times before wintering.And each treatment sample was taken back to the laboratory to determine the resistance related physiological and biochemical indexes,such as the membrane stability,osmotic adjustment,membrane lipid peroxidation,antioxidant enzymes in the leaves.The results shown that exogenous Ca2+had a significant impact on the resistance index of L.rotate after low temperature treatment in different concentrations.The degree of influence varies with the treatment concentrations and spray times.The proper CaCl2 concentration(the best concentration located to 2 twice 15 mmol·L-1)can significantly regulate soluble sugar content,antioxidant enzymes such as SOD and POD activities,which consequently mitigate the membrane lipid peroxidation and enhance the stability of the membrane systems.Moreover,exogenous Ca2+can reduce the relative conductivity and MDA content,and increase root activity.In conclution,exogenous Ca2+can increase the rate of re-green of L.rotata,and which have a potential application value in domestication and nurture of L.rotate.
