摘  要:目的研究算盘子不同极性萃取物的抑菌能力及最有效提取物对小鼠大肠杆菌性腹膜炎的治疗作用机制。方法采用不同极性溶剂(石油醚、二氯甲烷、乙酸乙酯、正丁醇、蒸馏水)对算盘子提取液进行萃取,得到5种提取物,测定各提取物对大肠杆菌和金黄色葡萄球菌的抑制效果,得到最有效的萃取方式。将50只小鼠随机分为正常组、模型组、小檗碱阳性组(0.0774g/ml)和高、低剂量组(0.1548、0.0774g/ml)。腹腔注射大肠杆菌建立大肠杆菌性腹膜炎模型,连续5天给药治疗。采用酶联免疫吸附法检测血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、肿瘤坏死因子α(TNF-α)、白细胞介素1(IL-1β)、γ干扰素(IFN-γ)的含量;取小肠、肝脏、肾脏进行HE染色并用免疫组织化学法检测小肠组织中金属基质蛋白酶9(MMP-9)、核因子κB(NF-κB)、C反应蛋白(CRP)的表达情况。结果算盘子乙酸乙酯提取物对大肠杆菌和金黄色葡萄球菌生长具有显著的抑制作用。与正常组比较,模型组小鼠SOD、GSH、IFN-γ水平显著降低(P<0.01),MDA、TNF-α、IL-1β水平显著升高(P<0.01),出现肠黏膜紊乱炎性细胞浸润,肝脏、肾脏重要器官炎症及损伤。与模型组比较,高、低剂量组能够有效降低血清中MDA及TNF-α、IL-1β的含量(P<0.05),提高SOD、GSH及IFN-γ的含量(P<0.05)。并能够降低CRP、MMP-9、NF-κB蛋白的表达水平(P<0.05),有效地减轻炎性反应并促进肠道、肝脏、肾脏组织修复。结论算盘子乙酸乙酯提取物具有治疗大肠杆菌性腹膜炎的作用,其机制可能与抑菌、抗炎、抑制机体氧化应激反应有关。Objective To investigate the bacteriostasis of different polar extracts from glochidion puberum in vitro,and to explore the most effective extracts for the treatment of E colil peritonitis in mice and its mechanism of action.Methods Five kinds of extracts were extracted with different polar solvents(petroleum ether,methylene chloride,ethyl acetate,n-butanol,distilled water)from Glochidion Puberum,and their inhibitory effects on escherichia coli and staphylococcus aureus were determined and most effective extraction method is obtained.Fifty mice were randomly divided into normal group,model group,berberine positive group(0.0774g/ml)and high and low dose groups(0.1548,0.0774g/ml).Establishment of E colil peritonitis model by intraperitoneal injection of Escherichia coli for 5 days.ELISA assay was adopted to determine the contents of MDA,SOD,GSH,TNF-α,IL-1β,IFN-γin serum;The intestine,liver and kidney were stained with HE and the expression of MMP-9,NF-κB and CRP in intestinal tissue was detected by immunohistochemistry.Results The ethyl acetate extract of glochidion puberum has significant inhibitory effect on the growth of Escherichia coli and Staphylococcus aureus.So it was used in animal experiment compared with blank control group,model control group could decrease the contents of SOD,GSH,and IFN-γin serum(P<0.01)significantly.The contents of MDA,TNF-α,IL-1βwere increased significantly(P<0.01).Suffered from significant disturbances of intestinal mucous membrane and flammatory cell infiltration;and induced inflammation and injury of liver and kidney.Compared with the model group,the high and low dose groups could decrease the contents of MDA,TNF-αand IL-1βin serum(P<0.05),increase the contents of SOD,GSH and IFN-γ(P<0.05).It can reduce the expression levels of CRP,MMP-9,NF-κB protein(P<0.05),effectively alleviate inflammation and promote tissue repair.Conclusion The ethyl acetate extract of Glochidion Puberum can treat E colil peritonitis may be relate to inhibiting the growth of bacteria,reducing inf
