摘  要:目的观察冬凌草甲素对雄激素缺乏诱导的大鼠骨质疏松症的影响。方法30只6个月龄SPF级SD大鼠随机分为睾丸切除+冬凌草甲素补充组(A组,n=10)、睾丸切除组(B组,n=10)和假手术组(C组,n=10)。3个月后采用双能X射线检测骨密度,ELISA检测血清骨代谢指标Ⅰ型胶原C端交联肽(CTX-1)、Ⅰ型原胶原N端前肽(P1NP),micro-CT检测骨微结构,并比较组间差异。结果与C组相比,B组CTX-1水平升高,P1NP水平降低,骨密度下降,骨微结构恶化(P<0.05);与B组相比,A组血清P1NP水平升高,CTX-1水平降低,骨密度增加,骨微结构修复(P<0.05)。结论冬凌草甲素能够有效预防雄激素缺乏诱导的大鼠骨质疏松症,修复骨微结构和增加骨密度,有效降低骨折风险。Objective To observe the effects of oridonin on androgen deficiency-induced osteoporosis in rats.Methods Thirty 6 months old SPF Sprague Dawley rats were randomly divided into three groups with 10 rats each.The rats in group A were treated with orchiectomy and oridonin supplementation,those in group B were treated with orchiectomy,and those in group C were sham operated.Three months later,bone mineral density was detected using dual energy X ray and serum bone metabolism index C-terminal cross-linking telopeptide of typeⅠcollagen(CTX-1)and N-terminal propeptide of typeⅠprocollagen(P1 NP)were detected by ELISA,and bone microstructure was examined by micro-CT.Results Compared with group C,group B had higher serum CTX-1 concentration,lower P1 NP concentration and lower bone density with deteriorated bone microstructure(P<0.05).Compared with group B,serum P1 NP level was increased,CTX-1 level was decreased,bone density was increased and bone microstructure was repaired in group A(P<0.05).Conclusion Oridonin supplementation may effectively prevent androgen deficiency-induced osteoporosis in rats,repair bone microstructure and enhance bone density,which benefits reducing the risk for fracture.
