摘 要:大豆胞囊线虫病(Soybean cyst nematode,Heterodera glycines Ichinohe)是一种严重影响大豆产量的病害,我国东北地区以3号生理小种(SCN Race 3)为主。QTL定位是挖掘抗性基因最有效的方法之一,与SCN相关的QTL报道较多,其由不同遗传群体获得,但在不同遗传群体间较难重复。研究采用图谱整合软件BioMercatorV4.2挖掘SCN Race 3抗性基因。将收集整理的10篇文献发表的33个QTL整合在遗传图谱GmComposite2003上,通过元分析方法得到一致性QTL,最终获得3个抗性候选区段,发现18个SCN抗性基因,其中17个PK型基因和1个LRR型基因,分别位于7、15和18染色体中。对潜在SCN抗性基因分析蛋白结构域发现,17个PK型基因分别含有丝/苏氨酸蛋白激酶活性、钙依赖蛋白激酶活性、分裂原活化蛋白激酶活性、半胱氨酸蛋白激酶和组/精/赖氨酸蛋白激酶活性。这些基因编码蛋白结构域与植物抗病相关,结果可为抗SCN基因挖掘和选育抗SCN品种(系)提供理论基础。Soybean cyst nematode(SCN,Heterodera glycines Ichinohe)is a serious disease affecting soybean yield.SCN Race 3 is dominant in northeast China.QTL mapping is one of the most effective methods for mining resistance genes;however,these QTLs were obtained from different genetic populations and are difficult to repeat among different genetic populations.In this study,map integration software BioMercatorV4.2 was used to mine SCN Race 3 resistance genes.The total 33 QTL published in 10 articles were integrated into the genetic map GmComposite2003.Additionally,the consistent QTLs were obtained by meta-analysis,and finally three resistance candidate segments were obtained,and 18 SCN resistance genes,17 PK type genes and 1 LRR type gene were found,which were located in 7,15 and 18 chromosomes.Protein domain analysis of potential SCN resistance genes showed that 17 PK genes contained serine/threonine protein kinase activity,calcium dependent protein kinase activity,mitogen activated protein kinase activity,cysteine protein kinase activity and group/arginine protein kinase activity.Combined with the previous research results,it was shown that the domain of proteins encoded by these genes was related to plant disease resistance,which provided a theoretical basis for SCN-resistant gene mining and breeding of SCN-resistant varieties(lines).
