摘 要:为探讨HMG box蛋白1(HMG box protein 1,HBP1)基因启动子区多态性对鸡腹脂率的影响,以东北农业大学高、低脂双向选择品系第十九世代肉鸡为试验材料,采用直接测序法检测HBP1基因多态性和个体基因型,共获得168个SNPs位点基因型。应用滑动窗口法对位于该基因启动子区54个多态性位点构建单倍型,分别开展单位点和单倍型与鸡腹脂率的关联分析。结果表明,HBP1基因启动子区有7个(g.-5335G>A、g.-4354T>A、g.-3328C>T、g.-3327G>A、g.-1013T>C、g.-990G>A和g.-159A>C)位点显著影响肉鸡腹脂率(P<0.05)。此外,等位基因频率分析显示,这些位点等位基因频率在两系间分布存在极显著差异(P<0.0001)。研究结果表明,HBP1基因为影响鸡腹脂率重要候选基因,7个多态性位点为影响鸡腹脂率重要分子标记。In order to investigate the effects of polymorphisms of HMG box protein 1(HBP1)gene on abdominal fat percentage in chickens,the19 th generation population of the chicken divergently selected for abdominal fat content of Northeast Agricultural University(NEAUHLF)was used in the present study.The direct sequencing was used for the detection of HBP1 gene polymorphisms and individual genotyping.A total of 168 SNPs were found.Fifty-four SNPs on the promoter region were applied to construct haplotypes using sliding window method,association of individual SNP genotypes and haplotypes with chicken abdominal fat percentage was analyzed,respectively.The results showed that there were seven loci(g.-5335 G>A,g.-4354 T>A,g.-3328 C>T,g.-3327 G>A,g.-1013 T>C,g.-990 G>A and g.-159 A>C)on the promoter of HBP1 gene that significantly affected abdominal fat percentage(P<0.05).In addition,analysis of SNP alleles showed that significant differences in allele frequencies of seven SNPs between two lines were observed(P<0.0001).The findings suggested that HBP1 gene could be an important candidate gene responsible for abdominal fat in chickens,and seven SNPs were important molecular markers for abdominal fat percentage in chickens.
