摘  要:目的探讨miR-3162-3p对卵清蛋白(OVA)致敏小鼠肺部炎症反应的影响及可能机制。方法选取24只小鼠,随机均匀分成对照组、对照+mimic组、致敏组、致敏+mimic组。致敏组采用腹腔注射OVA致敏,致敏+mimic组致敏后腹腔转染miR-3162-3p化学合成类似物mimic;对照组、对照+mimic组进行相应对照处理。各组采用瑞氏染色对肺泡灌洗液(BALF)进行白细胞分类计数,HE染色检测肺组织中炎症细胞浸润程度,ELISA检测肺组织炎症因子IL-4水平,qRT-PCR检测肺组织miR-3162-3p和靶基因β-catenin mRNA表达。结果与对照组、致敏组分别比较,对照+mimic组、致敏+mimic组小鼠BALF细胞总数增加、巨噬细胞比例升高、淋巴细胞比例降低,肺组织炎症细胞浸润程度增强、IL-4水平升高、β-catenin mRNA表达水平降低,差异均有统计学意义(P均<0.05)。结论miR-3162-3p可能通过降低肺组织靶基因β-catenin的表达水平从而加剧OVA致敏小鼠肺部炎症反应。Objective To investigate the effect of miR-3162-3p on pulmonary inflammation in ovalbumin(OVA)-sensitized mice and its possible mechanism.Methods Twenty-four mice were randomly divided into the control group,control+mimic group,sensitization group,and sensitization+mimic group.In the sensitization group,OVA was injected intraperitoneally to sensitize the mice.In the sensitization+mimic group,miR-3162-3p mimic,a chemosynthetic analogue,was transfected intraperitoneally into mice after OVA sensitization.In the control group and the control+mimic group,the corresponding control treatments were carried out.The leukocytes in bronchoalveolar lavage fluid(BALF)were counted by Wright's staining.The inflammatory cell infiltration in the lung tissues was detected by HE staining.The level of IL-4 in the lung tissues was detected by ELISA.The expression levels of miR-3162-3p and target geneβ-catenin mRNA in the lung tissues were detected by qRT-PCR.Results Compared with the control group and the sensitization group,the BALF cells increased,the proportion of macrophages increased,the proportion of lymphocytes decreased,the inflammatory infiltration in the lung tissues increased,the level of IL-4 increased,and the expression level ofβ-catenin mRNA decreased in the control+mimic group and the sensitization+mimic group(all P<0.05).Conclusion The miR-3162-3p could exacerbate pulmonary inflammation in OVA-sensitized mice through decreasing the expression of target geneβ-catenin in the lung tissues.
