摘 要:目的探讨舒脑欣滴丸(SNX)对大鼠脑膜微循环障碍的治疗作用。方法70只Wistar大鼠随机分为正常对照组、模型对照组、SNX 13mg/kg组、SNX 26mg/kg组、SNX 52 mg/kg组、脑心通和尼莫地平组各10只。各组分别采用相应药物连续灌服给药7 d,每天1次,末次给药后1 h,大鼠尾静脉注射15%高分子量右旋糖酐5 mL/kg,制备脑微循环障碍模型。监测各组不同时间点硬脑膜表面血流量,测定血浆NO、ET-1水平。结果与正常对照组比较,模型对照组注射15%高分子量右旋糖酐后5~120 min时脑膜微循环血流量减少(P均<0.01)。与模型对照组比较,造模后30~90 min时SNX 26mg/kg组、SNX 52 mg/kg组脑软脑膜血流量增加(P<0.05或<0.01);与模型对照组比较,SNX 52 mg/kg组大鼠血浆内皮因子NO水平升高,ET-1浓度降低(P<0.05或<0.01)。SNX 52 mg/kg与临床等效剂量的脑心通、尼莫地平作用相当。结论SNX能改善大鼠脑微循环障碍。Objective To investigate the effect of Shunaoxin dropping pills(SNX)on the meningeal microcirculation disturbance in rats.Methods Wistar rats(70 cases)were randomly divided into the control group,model group,13 mg/kg SNX group,26 mg/kg SNX group,52 mg/kg SNX group,Naoxintong capsule group,and nimodipine group.Rats were administrated continuously for 7 days,once daily.One hour after the last administration,the rats were injected with 15%high molecular weight dextran 5 mL/kg to prepare the cerebral microcirculation disturbance models.The blood flow on the surface of the dura mater was monitored at different time points in each group,and the levels of plasma NO and ET-1 were measured.Results Compared with the control group,the meningeal microcirculation blood flow decreased at 5 to 120 minutes after the injection of 15%high molecular weight dextran in the model group(all P<0.01).Compared with the model group,cerebral pia mater meningeal blood flow increased in the 26 mg/kg SNX group and 52 mg/kg SNX group at 5 to 120 minutes after modeling(P<0.05 or P<0.01).In the 52 mg/kg SNX group,the plasma endothelial factor NO level increased,and the ET-1 concentration decreased(P<0.05 or P<0.01).The effect of SNX(52 mg/kg)is equivalent to the effect of Naoxintong capsule and nimodipine in the clinically equivalent doses.Conclusion SNX can improve cerebral microcirculation disturbance in rats.
