摘 要:目的探讨腺苷预处理对大鼠脑缺血再灌注损伤后Caspase-3的表达及细胞凋亡的影响。方法将75只雄性Sprague-Dawley大鼠采用随机数字表法分为假手术组(Sham组)、缺血再灌注组(IR组)及腺苷预处理缺血再灌注组(AP组)各25只。AP组术前3 d腹腔注射腺苷注射液(剂量为1.5 mg/kg,生理盐水稀释至2 mL,1次/d);Sham组及IR组术前3 d腹腔注射等量生理盐水。线栓法制备左侧局灶性脑缺血/再灌注模型。缺血2 h再灌注24 h时,三组各随机抽取5只大鼠三组行TTC染色测定脑梗死体积。三组余20只分别取5只于缺血2 h再灌注6、24、48、72 h时,采用免疫组织化学法检测不同时刻Caspase-3的表达,采用TUNEL法检测不同时刻细胞凋亡数目。结果再灌注24 h,Sham组未见脑梗死病灶;与Sham组比较,IR、AP组脑梗死体积百分比增大(P均<0.05);与IR组比较,AP组脑梗死体积百分比减小(P<0.05)。与Sham组比较,IR组与AP组Caspase-3阳性细胞及凋亡阳性细胞表达均显著增多(P均<0.05);与IR组比较,AP组Caspase-3阳性细胞及凋亡阳性细胞表达明显减少(P<均0.05)。结论腺苷预处理能减轻脑缺血再灌注损伤,可能与抑制Caspase-3表达、减少细胞凋亡有关。Objective To observe the effects of adenosine preconditioning on the expression of Caspase-3 and apoptosis of rats after cerebral ischemia-reperfusion injury.Methods Seventy-five male Sprague-Dawley rats were randomly divided into the sham operation group(Sham group),ischemia-reperfusion group(IR group),and adenosine preconditioning ischemia-reperfusion group(AR group),with 25 rats in each group.The rats in the AP group received intraperitoneal injection of adenosine injection at 3 days before operation(with dose of 1.5 mg/kg,diluted to 2 mL with normal saline,once daily);the same amount of normal saline was intraperitoneally injected into the rats of the Sham group and the IR group at 3 days before operation.The left cerebral ischemia/reperfusion model was prepared by suture method.After 2-hour ischemia and 24-hour reperfusion,5 rats in each group were randomly selected for TTC staining to measure the volume of cerebral infarction.Five of the remaining 20 in the three groups were taken at 6,24,48,and 72 h after 2-hour ischemia and reperfusion,respectively,and the expression of Caspase-3 was detected by immunohistochemistry and the number of apoptosis was detected by TUNEL method.Results There was no cerebral infarction in the Sham group after 24 h of reperfusion;compared with the Sham group,the percentage of cerebral infarction volume in the IR and AP groups increased(both P<0.05);compared with the IR group,the percentage of cerebral infarction volume in the AP group decreased(P<0.05).Compared with the Sham group,the expression of Caspase-3 positive cells and apoptosis positive cells in the IR group and the AP group increased significantly(all P<0.05);compared with the IR group,the expression of Caspase-3 positive cells and apoptosis positive cells in the AP group decreased significantly(all P<0.05).Conclusion Adenosine preconditioning may alleviate cerebral ischemia-reperfusion injury by inhibiting Caspase-3 expression and decreasing the apoptosis.
