摘  要:目的探索黄芪多糖体外对人血管内皮细胞HUVECs诱导的胃癌细胞系SGC7901细胞EMT转化及其转移的影响。方法 MTT检测黄芪多糖对HUVECs、SGC7901细胞增殖抑制的影响。建立Transwell细胞共培养系统,并分为SGC7901组(单独培养SGC7901细胞)、HUVECs/SGC7901组(共培养HUVECs/SGC7901细胞)、黄芪多糖组(黄芪多糖干预共培养HUVECs/SGC7901细胞),Transwell实验检测各组SGC7901细胞侵袭能力,qRT-PCR检测各组细胞E-cadherin、Vimentin mRNA表达,免疫荧光检测各组细胞MMP-2、MMP-9蛋白表达,Western blot检测各组细胞LOX、Snail1蛋白表达。结果与单独培养的SGC7901细胞相比,共培养的SGC7901细胞侵袭能力,MMP-2、MMP-9、LOX、Snail1蛋白及Vimentin mRNA表达均升高(P<0. 01),E-Cadherin mRNA表达下降(P<0. 05)。经黄芪多糖干预后,共培养的SGC7901细胞侵袭能力、MMP-2、MMP-9、LOX、Snail1蛋白及Vimentin mRNA表达下降(P<0. 01),ECadherin mRNA表达升高(P<0. 05)。结论与HUVECs细胞共培养可激活SGC7901细胞EMT转化,促进SGC7901细胞转移,但黄芪多糖可通过抑制EMT转化阻止非接触共培养人血管内皮细胞诱导的SGC7901转移。AIM To investigate the effects of in vitro Astragalus polysaccharides( APS) on EMT transformation and metastasis of human gastric cancer cell line SGC7901 induced by HUVECs. METHODS MTT was used to detect the antiproliferative effect of APS on HUVECs and SGC7901 cells. After the establishment of Transwell cell co-culture system, cells were divided into SGC7901 group( single-cultured SGC7901 cells),HUVECs/SGC7901 group( co-cultured HUVECs/SGC7901 cells),and APS group( APS interfering co-culture of HUVECs/SGC7901 cells). Cells of each group were subjected to detection of the invasion ability of SGC7901 cells by Transwell;detection of the expression of E-cadherin and Vimentin mRNA by qRT-PCR;determination of the expression of MMP-2 and MMP-9 proteins by immunofluorescence;detection of the LOX,Snail1 protein expression by Western blot. RESULTS Compared with single-cultured SGC7901 cells,the co-cultured SGC7901 cells had increased invasion capacity,MMP-2,MMP-9,LOX,Snail1 protein,and Vimentin mRNA expression( P<0. 01),and decreased E-Cadherin mRNA expression( P < 0. 05). But these data may be altered with the intervention of APS as it brought forth reduced invasiveness,MMP-2,MMP-9,LOX,Snail1 protein and Vimentin mRNA expression( P<0. 01),and increased E-Cadherin mRNA expression( P<0. 05). CONCLUSION SGC7901 cells may achieve increased metastasis due to promoted activation of the EMT transformation by co-cultured HUVECs cells,but APS can reverse the alteration through inhibiting EMT transformation.
