摘  要:[目的]钝齿棒杆菌中有两个黄素还原酶,一个是由frd1编码的F181,另一个是由frd2编码的F188。该文分别对这两个基因进行了敲除,比较二者分别对精氨酸产量的影响。[方法]基因敲除采用的是无痕敲除技术。[结果]敲除frd1的菌株其最终的L-Arg产量达到21. 36 g/L,较出发株提高了10. 6%;葡萄糖消耗速率比出发株慢,但菌体生长量提高显著,提高了约33. 3%。敲除frd2摇瓶发酵产L-Arg的量较出发株提高了3. 15%;菌体生长量介于出发株及敲除frd1的菌株之间;葡萄糖消耗与敲除frd1的菌株相当;敲除frd1的菌株积累NADH最多,产生H2O2最低。[结论]敲除frd1比敲除frd2对提高精氨酸的产量效果显著。[Objective] Two kinds of flavin reductases exist in Corynebacterium crentum,one is F181 encoded by frd1 and the other is F188 encoded by frd2. In this paper,the two genes were knocked out,and the disruption effects on arginine production were compared.[Method] Markerless deletion technology was employed. [Result] The results showed that the final L-Arg yield of frd1-deleted strain reached 21. 36 g/L,which was 10. 6% higher than that of the starting strain;the glucose consumption rate was slower than that of the starting strain,but the growth of the cell was significantly increased,which was increased by about 33. 3%. The L-Arg production of frd2-deleted strain with shake flask was increased by 3. 15% compared with the starting strain;the growth of the cell was between the starting strain and the frd1-deleted strain;the glucose consumption was nearly equivalent to the frd1-deleted strain. The frd1-deleted strain can accumulate the highest concentration of NADH and the lowest concentration of H2O2.[Conclusion]Knockout frd1 was more effective in increasing arginine production than knockout frd2.
