摘 要:目的运用网络药理学方法对达原饮进行分析,探索达原饮治疗发热的相关靶点与通路,明确其作用机制与潜在治疗疾病,为后续研究提供指导。方法通过中药系统药理学数据库与分析平台(TCMSP)检索达原饮各组成药物的主要化学成分及作用靶点,参考类药五原则并结合口服生物利用度对成分靶点数据进行筛选,通过String数据库检索筛选后的靶点,得到达原饮作用靶点基因,在CTD数据库中检索发热(fever)相关作用靶点基因,对二者进行比较分析,取重复值,得到达原饮治疗发热相关靶点;在String数据库中检索达原饮治疗发热相关靶点,得到达原饮治疗发热相关通路;利用Cytoscape3.7.0软件对通路进行模块化分析,选择分值最高者,在WEB-based GEne SeT AnaLysis Toolkit中进行可视化分析。结果得到达原饮治疗发热相关靶点432个,相关通路75条,其中特异性疾病相关通路21条,非特异性疾病相关通路54条。结论达原饮治疗发热的作用机制涉及以代谢为中心的多种途径,可能通过多靶点复杂通路的共同干预起效。本研究就达原饮对于癌性发热的治疗补充了数据支持,并推测达原饮治疗神经退行性疾病具有可能性。Objective To explore the related targets and mechanism of Dayuan Decoction in treating fever by analyzing the prescription with the method of network pharmacology;To clarify the potential therapeutic diseases of Dayuan Decoction;To provide guidance for the follow-up study. Methods The main chemical components and action targets of the constituent medicine of Dayuan Decoction were retrieved through TCMSP. Component target data were screened based on the five principles of similar medicines and oral bioavailability. The screened targets were retrieved through String database to obtain the target genes of Dayuan Decoction. The disease(fever) was searched in the CTD to obtain the relevant target genes. The two were compared and analyzed, and the repeated values were taken to obtain the target related to the treatment of fever by Dayuan Decoction. The fever-related targets treated by Dayuan Decoction were retrieved in String, and fever-related pathway of Dayuan Decoction was obtained;Cytoscape 3.7.0 was used to perform a modular analysis of these pathways, the one with the highest score was selected, and a visual analysis was performed in the WEB-based GEne SeT AnaLysis Toolkit. Results Totally 432 related targets and 75 related pathways of Dayuan Decoction in treating fever were obtained, among which 21 were specific disease-related pathways and 54 were non-specific disease-related pathways. Conclusion The mechanism of Dayuan Decoction in treating fever involves multiple pathways centered on metabolism and the treatment is possibly achieved through joint intervention with multiple targets and complex pathways. This article supplemented the data support for Dayuan Decoction for the treatment of cancerous fever, and speculated that Dayuan Decoction has the possibility of treating neurodegenerative diseases.
